摘  要:目的考察补肝散醇提物不同极性提取部位的抗抑郁活性及初步作用机制。方法采用急性应激抑郁模型,小鼠分别灌胃补肝散醇提物(1.92 g/kg)、石油醚提取物A(0.244,0.122 g/kg)、二氯甲烷提取物B(0.160,0.077 g/kg)、乙酸乙酯提取物C(0.232,0.116 g/kg)、正丁醇提取物D(1.04,0.518 g/kg)及阳性对照药盐酸帕罗西汀(PX,26 mg/kg),连续给药14 d,测定各组小鼠在悬尾实验和强迫游泳实验中的不动时间;采用酶联免疫吸附测定法,考察活性提取部位B、C、D对小鼠大脑额叶皮质中神经递质5-羟色胺(5-HT)和多巴胺(DA)含量的影响。结果 PX、醇提物、提取物C、D及B高剂量均能显著缩短小鼠悬尾和游泳不动时间,提取物A及B的低剂量有缩短小鼠悬尾和强迫游泳不动时间的趋势,但与对照组比较无明显差别。提取物C、D及B的高剂量均能明显增加实验小鼠额叶皮质中单胺类神经递质5-HT含量,对DA含量有增加趋势。结论补肝散醇提物的抗抑郁活性部位主要集中于乙酸乙酯、正丁醇提取部位,其次为二氯甲烷提取部位;其抗抑郁的作用可能与增加脑内单胺类神经递质5-HT、DA含量有关。Objective To study the antidepressant effect and action mechanism of the different polar extractions of Bugansan.MethodsThe ethanol extract (1.92 g/kg) , the petroleum ether extract (A, 0.244 g/kg, 0.122 g/kg), the dichloromethane extract (B, 0.160 g/kg, 0.077 g/kg), the ethyl acetate extract (C, 0.232 g/kg, 0.116 g/kg) and the butyl alcohol extract (D, 1.04 g/kg, 0.518 g/kg) of Bugansan were given intragastrically to acute stress-induced depression mice for 15 days, respectively, with paroxetine hydrochloride (PX, 26 mg/kg) as positive control. The tail-suspension test and forced swimming test were used. The effect of different polar extractions (B, C, D) on the contents of serotonin (5-HT) and dopamine (DA) were investigated by enzyme-linked immunosorbent assay (ELISA).Results Compared with positive control, the forced swimming time and the tail-suspension time of the animals in ethanol extract, B, C and D treated groups were significantly shortened. The contents of 5-HT of the animals in B, C and D treated groups increased obviously. There was an increasing trend of DA of the animals in B, C and D treated groups. Conclusion The dichloromethane extract, ethyl acetate extract and butyl alcohol extract of Bugansan can be the active parts with antidepressant effect. The antidepressant effect may be related to the increasing contents of 5-HT and DA.
