摘 要:稀土上转换发光纳米材料在生物领域应用的前提是具有亲水性和可以偶联蛋白的官能团。先将十六烷基三甲基溴化铵(CTAB)中的碳链通过疏水作用插入到溶剂热法制备的NaYF4:Yb,Er纳米粒子表面油酸分子中,然后通过多巴胺(DA)在其表面氧化聚合形成具有氨基的亲水性纳米粒子(UCNPs@DA)。分别采用透射电子显微镜(TEM)、傅里叶变换红外光谱(FT-IR)、光纤光谱对其形貌、表面官能团和上转发光性能进行了表征与分析。考察了在修饰过程中DA的质量浓度、滴加速度等因素对形成UCNPs@DA的影响。结果表明,当DA质量浓度为0.20 mg/mL、滴加速度为1.25 mL/min时,试样具有较好水中分散性及发光效率。UCNPs@DA在540 nm处的发光强度是未修饰试样的70%,而650 nm处的光强未发生衰减。UCNPs@DA有望成为生物检测中很好的发光标志物。Good hydrophilicity and appropriate functional group are the prerequisites for bio-applications of up-conversion nanoparticles (UCNPs).The long carbon chain of hexadecyl trimethyl ammonium bromide ( CTAB) was inserted into the oleic acid molecules on the surface of solvothermally synthesized NaYF4:Yb, Er through hydrophobic interaction.This activated NaYF4:Yb,Er nanoparticles were then converted to water soluble and amine group bearing one (UCNPs@DA) through oxidative polymerization of dopamine (DA). The morphologies,surface functional groups and luminescence properties of NaYF4:Yb,Er samples were characterized by transmission electron microscope (TEM), Fourier transform infrared spectrometer (FT-IR) and upconversion luminescence spectrum. The effects of DA solution mass concentration and its dropping speed were investigated in details. UCNPs@ DA with good hydrophilicity and luminescence property was obtained using a 0?20 mg/mL DA solution at a dropping speed of 1.25 mL/min. After DA modification,about 70% of original luminescence at 540 nm was preserved while the light intensity at 650 nm remained unchanged.The UCNPs@DA could act as a good label for biological detection.
