摘  要:目的探讨Goldeneye^(TM) DNA身份鉴定系统25A试剂盒在亲子鉴定中的应用价值。方法对738例亲子鉴定的1 680份样本,采用Goldeneye 25A试剂盒复合扩增23个常染色体STR基因座和1个Y-STR基因座,评估该系统在亲子鉴定中的排除能力和突变率。结果采用Goldeneye 25A系统,累积非父排除率大于0.999 999 999 999 999999 999 999 999 999。738例亲子鉴定案例中排除亲子关系63例,最常见为9~13个基因座排除;等位基因突变事件22例,其中一步突变21例,二步突变1例,20例表现为1个STR基因座突变,1例表现为2个STR基因座突变;D19S253与FGA基因座的突变率最高,均为5.42×10-3,父系突变与母系突变比例为6.33∶1。Y-STR与Amelogenin基因座的分型结果相符。结论 Goldeneye 25A系统用于亲子鉴定是高效的。Objective To assess the feasability of applying GoldeneyeTM DNA ID 25 A Kit in paternity test cases. Method 23 autosomal STR loci and 1 Y-STR locus of 1680 samples from 738 cases were amplified with Goldeneye 25 A kit to validate its exclusion power and mutation rate. Results The cumulated chance of exclusion and the cumulated discriminating power of Goldeneye 25 A were greater than 0.999,999,999,999,999,999,999,999,999,999. 63 out of 738 cases were excluded,most of which had 9-13 exclusion loci. 22 mutation events were observed. Among them, 21 mutations were one step and 1mutation was two steps; 20 cases showed single-locus mutation and 1 case showed two-loci mutation. D19S253 and FGA had the highest mutation rate of 5.42 ×10-3. The ratio of paternal mutation versus maternal mutation was 6.33 ∶ 1. The genotype results of Y-STR were consistent with those of Amelogenin. Conclusion Goldeneye 25 A is highly effective for paternity testing.
