摘  要:为探讨缬草(Valeriana officinalis L.)黄酮纯化工艺,以树脂对黄酮的吸附率和解吸率为评价指标,通过静态吸附-解吸试验对6种树脂进行优选,在此基础上开展动态吸附-解吸试验,对上样与洗脱条件进行优化。结果表明,采用聚酰胺柱层析纯化缬草黄酮,在上样液浓度2.0 mg/m L、上样流速1.5 m L/min、上样量20.0 m L、上样液p H 5.5的条件下,用70.0%乙醇洗脱,洗脱流速2.0 m L/min、洗脱剂70.0 m L纯化3次,洗脱液经干燥、检测,发现黄酮纯度由粗品17.58%提高到79.17%。该工艺简单、安全、环保,可得到较高纯度的黄酮,适合纯化缬草黄酮。In order to investigate the purification process of flavonoids from Valeriana officinalis L.,the optimum resin was chosen from 6 kinds of resins by the static absorption-desorption tests and the adsorption and desorption rate of resins to flavonoids were used as index. Based on the above results,the sample and elution conditions were optimized by the dynamic absorption-desorption experiments. Flavonoids from Valeriana officinalis L. were purified by polyamide column chromatography and the optimum sample conditions were concentration 2.0 mg/mL,velocity 1.5 mL/min,amount 20.0 mL and pH value 5.5,the optimal elution conditions were ethanol concentration 70.0%,velocity 2.0 mL/min and amount 70.0 mL. Under the above conditions,the purity of flavonoids was from 17.58% up to 79.17% by three times purification. The above approach is appropriate to purify flavonoids from Valeriana officinalis L. for it is safe,friendly to environment,easy to operate and can gain flavonoids safely with high purity.
