摘  要:目的:研究猪胎盘水提物(PPE)对小鼠的抗氧化及学习记忆调节作用。方法:将经D-半乳糖诱导成功的32只衰老小鼠随机分成PPE高、低剂量组、阳性对照组、模型组,分别以PPE 30,20 mg·kg^(-1)·d^(-1)、维生素E 20 mg·kg^(-1)·d^(-1)、0.9%Na Cl溶液20 m L·kg^(-1)·d^(-1)经皮给药,另取8只正常小鼠作为正常对照(0.9%Na Cl溶液20 m L·kg^(-1)·d^(-1)),连续给药6周。给药后每周称量小鼠体重。停药前1周开始采用Morris水迷宫法检测小鼠学习记忆能力。第6周末处死小鼠,取腹部皮肤及眼球取血,测量超氧化物歧化酶(SOD)、丙二醛(MDA)、羟脯氨酸(Hyp)的含量,并观察病理切片。结果:与正常组相比,模型组小鼠水迷宫找到平台所需时间明显延长,小鼠在平台象限内停留时间、停留时间百分比、穿越平台次数均显著降低(P<0.05)。模型组小鼠体重、SOD含量、MDA含量、Hyp含量较正常组均有显著差异(P<0.01,P<0.05),皮肤表皮层不规则,真皮层皱缩,胶原纤维排列紊乱无序。经过PPE治疗后,高、低剂量组小鼠水迷宫找到平台时间明显缩短,小鼠在平台象限内停留时间、停留时间百分比、穿越平台次数均增高;高、低剂量组的体重、SOD含量、Hyp含量均明显上升(P<0.01,P<0.05),MDA含量明显下降(P<0.01,P<0.05);PPE高、低剂量组小鼠的皮肤结构较完整,表皮层规则,真皮层厚度增加,胶原纤维排列整齐。结论:猪胎盘水提物具有抗氧化及改善小鼠记忆力的作用。Objective: To study the effects of pig's placental extract( PPE) on learning and memory abilities and antioxidative function in the model of mouse aging. Methods: Totally 32 aging mice induced by D( +)galactose were divided into PPE high,low dose groups,positive control group and model group and percutaneously administered with pig placental extract 30,20 mg·kg^(-1)·d^(-1),vitamin E 20 mg·kg^(-1)·d^(-1),or physiological saline20 m L·kg^(-1)·d^(-1),respectively. Another 8 healthy mice were assigned to the normal control group( NC group,physiological saline 20 m L·kg^(-1)·d^(-1)). All the mice were treated consecutively for 6 weeks. During the treatment,the body weight was measured every week. Morris water maze was used to test the learning and memory abilities of mice at the end of week 5. At the end of week 6,abdominal skins were collected to determine the contents of SOD,MDA,and Hyp and observe the pathological sections. Results: Compared with the NC group,the time required to find the platform( latency) was longer in the model group( P〈0. 05),the time and percentage of staying in platform quadrant and frequency of crossing the platform were decreased( P〈0. 05). The body weight and thecontents of SOD and Hyp were significantly lower than those in the NC group( P〈0. 01,P〈0. 05) and the MDA content was significantly higher than that in the NC group( P〈0. 01,P〈0. 05),with significant pathological changes. The epidermal layer of the skin of the mice was irregular,the thicknesses of the dermal layer was decreased significantly,and the collagen fibers in the model group were thinner and looser. After the intervention with PPE,the latency was shortened( P〈0. 05),and the time and percentage of staying in platform quadrant and frequency of crossing the platform were increased( P〈0. 05).
