摘  要:目的:研究淫羊藿苷(icariin,ICA)对氧化低密度脂蛋白(oxidized low density lipoprotein,oxLDL)诱导的巨噬细胞凋亡的影响,并探讨其机制。方法:体外培养巨噬细胞RAW264.7,给予ICA(10,20,40μmol·L^(-1))或5 mmol·L^(-1)4-苯丁酸(4-phenylbutyric acid,4-PBA)预处理1 h,然后加入100 mg·L-1ox LDL继续培养24 h,采用MTT法检测细胞活力;油红O染色检测泡沫细胞的形成;Annexin V-FITC/PI和比色法检测细胞凋亡和caspase-3活性,Western blot检测GRP78的表达水平。结果:与ox LDL组相比,ICA预处理组减轻ox LDL诱导的巨噬细胞的损伤,表现为细胞活力的增加,凋亡率下降,caspase-3活性降低,与内质网抑制剂4-PBA相似,降低内质网应激标志蛋白GRP78表达。结论:ICA可能通过抑制ox LDL诱导的过强内质网应激,降低caspase-3活性,抑制ox LDL诱导的细胞凋亡。Objective: To investigate the inhibitory effects of icariin( ICA) on oxidized low density lipoprotein( ox LDL)-induced macrophage apoptosis and the underlying mechanisms. Methods: RAW264. 7macrophages were pretreated with ICA( 10,20,40 μmol·L-1) or 5 mmol·L-14-phenylbutyric acid( 4-PBA) for 1h,then treated with ox LDL( 100 mg·L-1) for 24 h. The cell viability and apoptosis rate were detected by MTT assay and annexin-FITC / PI double staining method,respectively. Intracellular lipid droplets were assayed by oil red O staining,caspase-3 activity was measured by chromatometry,and endoplasmic reticulum stress markers( GRP78) was detected by Western blot. Results: Pretreatment with ICA( 20,40 μmol·L-1) could protect macrophages from injury induced by ox LDL,as shown by increased cell viability and decreased apoptotic rate and significantly attenuated lipid accumulation in RAW264. 7 cells. Compared with ox LDL group,the activity of capase-3 and the protein level of GRP78 in the macrophages were decreased in ICA pretreatment group. Conculsion:ICA can prevent macrophages damage induced by ox LDL,and the mechanisms may be partially involved with itsability to decrease the activity of caspase-3 and down-regulate endoplasmic reticulum stress.
