三种针法对PCPA失眠大鼠海马5-HT、5-HIAA含量的影响

摘　　要：目的:观察针刺失眠方、脐内环穴及失眠方合脐内环穴对PCPA失眠大鼠海马5-HT、5-HIAA含量影响,并对3种针法优选。方法:70只大鼠经随机、PCPA失眠造模及筛选后共分6组各10只,正常组、模型组无特殊处理,3个观察组分别给予针刺失眠方、针刺脐内环穴、针刺失眠方合脐内环穴干预,非穴组给予针刺非穴处理,治疗6 d后取大鼠海马匀浆,ELISA法检测5-HT和5-HIAA含量。结果:PCPA模型大鼠海马5-HT和5-HIAA明显降低,3种针法均显著升高5-HT、5-HIAA含量;而失眠方合脐内环穴针刺作用更好,优于2种单一方法并显示其叠加作用。结论:3种针法可能通过升高海马内5-HT、5-HIAA含量而改善失眠,实验支持失眠方合脐内环穴针刺为失眠症的优选干预方法。Objective： To observe the three acupuncture methods on affecting the content of 5-HT, 5-HIAA in hippocampal of PCPA insomnia model rats of the acupuncture of umbilical inner acupoints, the acupoint-prescription of insomnia, their combined method of the acupoint-prescription of umbilical inner acupoints and the insomnia acupointprescription, and to select the optimal acupuncture method. Methods： After being randomed, PCPA insomnia modeled, insomnia screened on the 70 male rats, the resulted 60 rats were randomly divided into six groups, and each group were 10. The normal group and the model group were given no special treatment, the three observation group were given acupuncture treatment of the umbilical inner acupoints, the acupoint-prescription of insomnia, their combined method of the acupoint-prescrlption of umbilical inner acupoints and the insomnia acupoint-prescription respectively. The non-acupoint group was dealed with acupuncture non-acupoint. After six days of treatment, in the hippocampus homogenates of the rats, the content levels of 5-HT and 5-HIAA were detected by enzyme-linked immunoassay. Results ： The contents of 5-HT, 5- HIAA in hippocampal of PCPA insomnia model rats were significantly reduced. Three acupuncture methods can improve the 5-HT and 5-HIAA contents in hippocampal of rats. Their combined method of the acupoint-prescription of umbilical inner acupoints and the insomnia acupoint-prescription displayed a better effective increasing the content of 5-HT, 5-HIAA than each single method of acupuncture and showed an additive effect. Conclusion： Three acupuncture methods might take affect on insomnia by the mechanism of improving the contents of 5-HT and 5-HIAA in hippocampal. The experiment suggested that the combined method of the acupoint-prescription of umbilical inner acupoints and the insomnia acupoint-prescription therefore was the preferred acupuncture.
