摘 要:目的研究宫内暴露于七氟醚对仔鼠海马CA1区神经元凋亡及早期认知功能的影响。方法成年性成熟Wistar雌鼠交配妊娠后取孕鼠8只,随机分为对照组七氟醚组,每组4只。七氟醚组于孕19天时暴露于2.4%七氟醚6h,对照组不给予任何药物。应用免疫组化法观察两组仔鼠在出生后第1天(PN1)、第14天(PN14)、第21天(PN21)海马CA1区caspase-3的表达情况。出生后32~37天对仔鼠进行Morris水迷宫实验以测定其认知功能。结果出生后第1天,七氟醚组仔鼠海马CA1区caspase-3总光密度值(IOD)明显高于对照组仔鼠[(60309.6±6403.6)比(17680.5±9326.7),P〈0.05],出生后第14、21天,七氟醚组仔鼠海马CA1区caspase-3总光密度值与对照组仔鼠相比,差异无统计学意义[(3213.7±582.8)象素比(8503.2±2402.5)象素,(2046.0±62.7)象素比(4839.9±716.3)象素,P〉0.05]。七氟醚组仔鼠逃避潜伏期与对照组仔鼠相比,差异无统计学意义[(80.0±13.5)s比(80.7±19.1)s,(52.2±10.3)s比(41.0±19.7)s,(29.2±11.4)s比(28.5±15.3)s,(22.0±5.5)s比(19.2±6.7)s,(16.2±2.8)s比(20.3±4.8)s,P〉0.05],七氟醚组仔鼠穿越目标象限次数与对照组仔鼠相比无统计学差异[(8.3±0.8)次比(8.2±3.0)次,P〉0.05]。结论孕晚期Wistar母鼠暴露于2.4%七氟醚对仔鼠海马CA1区神经元发育的影响呈"一过性损伤",并不会引起子代早期认知功能异常。Objective To investigate the effect of sevoflurane exposure in the uterus on the rat offspring's brain neural apoptosis in the CA1 region of hippocampus and the cognitive function at early age.Methods Eight adult female Wistar rats got pregnant.With the random number table method,pregnant rats were divided into2 groups:control group and sevoflurane group,with 4 rats in each group.Rats in sevoflurane group were exposed to 2.4% sevoflurane for 6 h on the gestational 19 th day and rats in control group were fed in the same condition in the absence of sevoflurane.The expression of caspase-3 in the CA1 region of hippocampus on PN,PN14 and PN21 were determined by immunohistochemistry.The early cognitive function of the offspring from PN32 to PN37 was tested by Morris water maze test.Results Compared to the offspring of control group,the offspring of sevoflu rane group had higher integral optical density(IOD) of caspase-3 in the CA1 region of hippocampus on PN1(60309.6±6403.6 vs 17680.5±9326.7,P〈0.05) and similar IOD on PN14 and PN21(PN14:3213.7±582.8 vs 8503.2±2402.5;PN21:2046.0±62.7 vs 4839.9±716.3;all P〈0.05).No significant differences in escape latency(80.0±13.5s vs80.7±19.1s,52.2±10.3s vs 41.0±19.7s,29.2±11.4s vs 28.5±15.3s,22.0±5.5s vs 19.2±6.7s,16.2±2.8s vs 20.3±4.8s)and explored number in the target quadrant(8.3±0.8 times vs 8.2±3.0 times) were noted between control group and sevoflurane group(all P〈0.05).Conclusion The expose to 2.4% sevoflurane during late pregnancy causes temporary injury to brain neural development in the CA1 region of hippocampus of rat offspring.It has no significant effect on the cognitive function of the rats at the early age.
