摘 要:目的研究稳心颗粒(WXKL)对血管紧张素Ⅱ(AngⅡ)诱导H9C2细胞肥大模型骨架蛋白的影响。方法体外培养H9 C2细胞,经血管紧张素Ⅱ诱导复制心肌细胞肥大模型,加入5 g/L稳心颗粒后,观察细胞的形态变化以及对骨架蛋白的影响。结果经AngⅡ诱导的H9C2心肌细胞形态发生改变,细胞横向拉伸,肿胀变大,细胞体积较正常心肌细胞增大;加入稳心颗粒后H9 C2心肌细胞形态有所恢复,但还是比正常细胞横向宽度增加。应用激光共聚焦显微镜下观察细胞形态发现,正常H9 C2细胞经10-7mol/L AngⅡ刺激48 h后,细胞形态明显肿胀变大,细胞的长度和宽度分别由(120.32±8.36)μm和(51.82±8.62)μm增加到(182.67±7.81)μm和(92.22±7.56)μm。加入稳心颗粒作用24 h后,AngⅡ组的H9C2心肌细胞形态有所恢复,细胞的长度和宽度为(160.35±5.66)μm、(55.87±4.52)μm。结论稳心颗粒在AngⅡ诱导的肥厚心肌H9C2细胞模型中,能改善细胞骨架蛋白,起到一定的改善心肌肥厚的作用。Objective To study the effect of Wenxin granule( WXG) on the cytoskeleton protein of angiotensin Ⅱ( AngⅡ) induced H9 C2 cell hypertrophy model. Methods H9 C2 cells were cultured in vitro,the model of cardiomyocyte hypertrophy was induced by AngⅡ. After dosing WXG with 5 g / L,the morphological changes of the cells and the cytoskeleton were observe. Results The morphological of H9 C2 cell induced by AngⅡ had changes,cell transverse tensile,swelling becomes larger,cell volume vs normal cells. After with the addition of WXG,H9 C2 myocardial cell morphology had been restored,but still larger than the transverse width of normal cells. And the discovery of cell morphology were observed under confocal laser scanning microscopy( CLSM),cell morphology was apparent swelling becomes larger,the length and the width of the cell respectively by 120. 32 μm ± 8. 36 μm and 51. 82 μm ± 8. 62 μm increased to 182. 67 μm ± 7. 81 μm and 92.22 μm ± 7. 56 μm. After 24 hours of WXG,H9 C2 myocardial cell morphology in AngⅡ group had been restored,the length and the width of the cell to 160. 35 μm ± 5. 66 μm,55. 87 μm ± 4. 52 μm. Conclusion In H9 C2 cell model of hypertrophic myocardium induced by AngⅡ,the WXKL can improve the cytoskeletal protein,which plays a role in improving cardiac hypertrophy.
