摘 要:为了筛选对多重耐药铜绿假单胞菌具有抑菌作用的中草药,并筛选铜绿假单胞菌的群体感应蛋白LasR的抑制剂,本实验利用纸片扩散法对59株临床分离的铜绿假单胞菌和1株PAO1进行抗生素药敏试验,并对其中3株菌进行中草药提取物体外抑菌试验,通过检测报告质粒中β-半乳糖苷酶的活性来筛选LasR蛋白抑制剂。实验发现,丁香、赤芍等中药粗提物具有良好的抑菌效果,部分中草药可通过影响LasR蛋白的水溶性或信号分子与受体蛋白的结合来实现对细菌群体感应的调控。The aim of this study is to screen the bioactive molecules from Chinese medicine,which either interfere the bacterial quorum sensing in multi-drug resistant Pseudomonas aeruginosa or simply inhibit its growth. The P. aeruginosa strains isolated from clinic samples were tested for antimicrobial sensitivity using disk diffusion method. Screening of quorum sensing inhibitors from Chinese herbal medicines were carried out by monitoring the activity of β-galactosidase( Lac Z) in reporter strains,which can be used for screening quorum-sensing signal molecules and inhibitors. The agar diffusion test results indicated that the alcoholic extracts of Clove and Radix Paeoniae Rubra showed strong antimicrobial activity against P. aeruginosa. The SDS-PAGE gel results further suggested thatthe active components of Chinese herbal medicine interfered bacterial quorum sensing by reducing the amount of active LasR protein in soluble fraction or competitively binding to the signal molecules binding domain in receptor proteins.
