摘  要:为观察铁皮石斛多糖对NO、cGMP、Na+-K+-ATP酶活性及心肌细胞动作电位的影响并探讨其效应机制,将清洁级Wistar大鼠分为7组,每组10只。模型组高脂饲料及寒冷刺激6周,对照组基础饲料常态饲养。模型组第35天~第42天皮下多点注射垂体后叶素(10μg/kg),对照组给等体积的生理盐水,每天1次。低、中、高剂量组,阿托品组和小檗碱组第35天~第42天分别灌胃铁皮石斛多糖(100、200、400mg/kg)、静脉注射阿托品(5mg/kg)、静脉注射小檗碱(8mg/kg),每天1次。第43天取材,检测血清NO、cGMP、Na+-K+-ATP酶活性及心肌细胞动作电位。结果显示,与对照组比较,模型组、小檗碱组血清NO、cGMP含量均显著升高(P<0.01);Na+-K+-ATP酶活性显著降低(P<0.01),APD、APD90、ERP均延长(P<0.05或P<0.01),模型组造模后、小檗碱组给药后心率均显著降低(P<0.01)。与模型组比较,高剂量组及阿托品组NO、cGMP含量均降低(P<0.05或P<0.01);Na+-K+-ATP酶活性升高(P<0.05或P<0.01),高剂量组及阿托品组APD、APD90、ERP均缩短(P<0.05或P<0.01),高剂量组及阿托品组心率显著增高(P<0.01)。表明铁皮石斛多糖对NO、cGMP、Na+-K+-ATP酶活性均有影响,与剂量有关,通过NO-GC-cGMP通路提高心率,同时也可能通过NO间接调节Na+-K+-ATP酶活性或直接调节Na+-K+-ATP酶活性而发挥作用。To observe the effects of Dendrobium candidum polysaccharides (DCP) on NO- GC- cGMP channel in rats with chronic cardiac arrhythmias of coronary heart disease and discuss its mechanisms, 70 clean grade Wistar rats were randomly divided into control group, model group, DCP low-dose group, medi- um-dose group,high-dose group,atropine group and berberine group 7 groups with 10 in each group. The rats in model group given were with high fatty diet and cold circumstance for 6 weeks while control group with ordinary diet and circumstance. From the 35th day to the 42nd day,the rats in model group was given were ih with pitutrin (10 μg/kg) while control group with equal normal saline. Based on model group,from the 35th day to the 42nd day, the rats in low-dose group,medium-dose group, high-dose group, atropine group and berberine group given were respectively ig with DCP(100 mg/kg,200 mg/kg, 400 mg/kg),iv with atropine (5 mg/kg)and or iv with berberine(8 mg/kg) ,once a day. On the 43rd day to the contents of serum NO,cGMP and activity of Na+-K+-ATP enzymes,AP of myocardial cells were examined. The re- sults showed that compared with control group, the rats in model group and berberine group, contents of serum NO and cGMP increased significantly (P〈0.01), activity of Na+-K+-ATP enzymes reduced significantly (P〈0.01). APD, APD90, ERP all increased (P〈0.05 or P〈0.01), heart rate reduced significantly (P〈0.01). Compared with model group,the rats in high-dose group and atropine group,contents of serum NO and cGMP reduced (P〈0.05 or P〈0.01) ,activity of Na+-K+-ATP enzymes increased (P〈0.05 or P 〈0.01), APD, APD90, ERP all reduced (P〈0. 05 or P〈0. 01), heart rate increased significantly (P〈 0.01). It was supposed that DCP took an effect on content of serum NO,cGMP and activity of Na+-K+- ATP enzymes to increase heart rate by NO- GC- cGMP channel,which is relevant to its dose.
