摘  要:目的研究罗格列酮对胰岛素抵抗(IR)3T3-L1脂肪细胞脂联素、瘦素及抵抗素的分泌及mRNA表达的影响。方法将3T3-L1脂肪细胞分为5组:A组为对照组,B、C、D、E组为不同水平罗格列酮处理的IR组,分别给予终浓度为0、0.1、0.5、1.0μmol/L的罗格列酮。检测各组细胞培养液的葡萄糖消耗量,分别采用酶联免疫吸附试验(ELISA)及实时荧光定量聚合酶链式反应(Q-RT-PCR)检测瘦素、脂联素及抵抗素的分泌及mRNA表达水平。结果与B组比较,D、E组细胞分泌的瘦素及抵抗素水平及其mRNA表达水平均降低,分泌的脂联素及其mRNA表达水平与葡萄糖消耗量均升高,差异均有统计学意义(P<0.05)。B组与C组细胞分泌的瘦素、脂联素及抵抗素水平比较,差异均无统计学意义(P>0.05)。结论罗格列酮可以通过调节瘦素、脂联素及抵抗素的表达及分泌减轻脂肪细胞的IR。Objective To study the influence of rosiglitazone on the secretion and mRNA expression of adiponectin,leptin and resistin in 3T3-L1 adipocytes with insulin resistance(IR). Methods 3T3-L1 adipocyteg were divided into 5 groups:group A was the control group,group 13,C, D and E were the IR groups treated with different concentrations of rosiglitazone. The group B, C,D and E were given 0 ,0. 1,0.5,1.0 μmol/L rosiglitazone, respectively. The consumption of glucose was detected, while the secretion and mRNA expression of leptin, adiponectin and resistin were detected by using ELISA and Q-RT-PCR respectively. Results Compared with the group B,the secretion and mRNA expression of leptin and resistin in group D and E were decreased, while the secretion and mRNA expression of adiponectin and the consumption of glucose were increased, these differences were sta- tistically signifieant(P〈0.05). The secretion of leptin, adiponectin and resistin had no significant difference between group B and group C(P〉0.05). Conclusion Rosiglitazone could alleviate the IR in adipocytes by regulating the expression and section of leptin, adiponectin and resistin.
