摘 要:目的:建立华中五味子不同部位中10种金属元素含量的测定方法。方法:采用氢化物-原子吸收光谱法(HG-AAS)测定药材中砷(As)、铅(Pb)、汞(Hg)的含量;采用火焰原子吸收光谱法(FAAS)测定药材中镉(Cd)、铜(Cu)、锰(Mn)、锌(Zn)、铁(Fe)、镁(Mg)、钙(Ca)的含量。结果:As、Pb、Hg、Cd、Cu、Mn、Zn、Fe、Mg、Ca检测质量浓度线性范围分别为0.000 12~0.015μg/ml(r=0.999 8)、0.000 10~0.002 5μg/ml(r=0.999 4)、0.000 068~0.005μg/ml(r=0.999 3)、0.000 28~0.040μg/ml(r=0.998 7)、0.009 8~2.0μg/ml(r=0.999 8)、0.009 6~10.0μg/ml(r=0.999 6)、0.002 4~5.0μg/ml(r=0.999 8)、0.098 7~5.0μg/ml(r=0.999 5)、0.001 8~1.5μg/ml(r=0.999 6)、0.035 8~20.0μg/ml(r=0.999 7);精密度、稳定性、重复性试验的RSD≤5.4%;加样回收率在92.5%~107.9%范围内,RSD≤5.1%(n=9)。结论:该方法操作简便、灵敏、准确,可用于华中五味子不同部位中10种金属元素的含量测定。华中五味子藤茎中Cu、Mn、Zn、Ca含量均高于果实;样品中亦检出微量As、Pb、Hg、Cd,但均未超标。OBJECTIVE: To establish a method for contents determination 10 kinds of inorganic elements in different parts of Schisandra sphenanthera. METHODS: Hydride generation atomic absorption spectrometry(HG-AAS) was used to determine the contents of As, Pb and Hg; flame atomic absorption spectrometry (FAAS) was used to determine the contents of Cd, Cu, Mn, Zn, Fe, Mg and Ca. RESULTS: The linear range was 0.000 12-0.015 μg/ml for As(r=0.999 8), 0.000 10-0.002 5μg/ml for Pb (r=0.999 4), 0.000 068-0.005 pg/ml for Hg(r=0.999 3),0.000 28-0.040 μg/ml for Cd(r=0.998 7),0.009 8-2.0 μg/ml for Cu(r= 0.999 8) ,0.009 6-10.0μg/ml for Mn(r=0.999 6), 0.002 4-5.0 μg/ml for Zn(r=0.999 8),0.098 7-5.0 μg/ml for Fe (r=0.999 5), 0.001 8-1.5 μg/ml for Mg(r=0.999 6) and 0.035 8-20.0 μg/ml for Ca(r=0.999 7);RSDs of precision, stability and reproducibility tests were no higher than 5.4% ; recovery was 92.5%-107.9% (RSD≤5.1% , n=9). CONCLUSIONS: The method is simple, sensitive, accurate, and can be used for the contents determination 10 kinds of inorganic elements in different parts of S. sphenanthera. The contents of Cu, Mn, Zn, Ca in vine and stem are higher than that in the fruits; As, Pb, Hg and Cd are also detected in sample, but the detected contents are all in the limit of the standard.
