补肾疏肝方对小鼠Lewis肺癌的抑制作用及相关机制研究

摘　　要：目的:观察补肾疏肝方对Lewis肺癌生长和转移的抑制作用并探讨其可能存在的机制。方法:选取模型复制成功的C57BL/6小鼠40只,随机分为模型组、顺铂组、中药组、综合组。模型组给予生理盐水灌胃0.4 m L·d-1;顺铂组在第1天、第3天、第5天给予顺铂腹腔注射0.5 m L(含顺铂0.1 mg);中药组给予中药灌胃0.4 m L·d-1(含生药0.7 g);综合组给予每日中药灌胃(同中药组)同时腹腔注射顺铂(同顺铂组),共干预21 d。第22天处死,剥取肿瘤称质量,计算抑瘤率;计算自发性肺转移灶数,计算肺转移抑制率;用免疫组织化学的方法检测瘤组织微血管密度(microvessel density,MVD)、血管内皮生长因子(vascular endothelial growth factor,VEGF)及基质金属蛋白酶-9(matrix m etalloproteinase-9,MMP-9)的表达。结果:与模型组相比,各用药组均能显著抑制肿瘤生长,差异有统计学意义(P<0.01);与中药组比较,综合组平均瘤质量显著降低(P<0.01)。与模型组相比,各用药组均能显著抑制肿瘤的自发性肺转移(P<0.05);与顺铂组和中药组比较,综合组自发性肺转移灶数显著降低(P<0.05)。与模型组相比,各用药组MVD、VEGF均显著降低(P<0.05);与顺铂组相比,综合组MVD、VEGF降低明显(P<0.05)。与模型组相比,各用药组MMP-9表达显著降低(P<0.01);与顺铂组和中药组相比,综合组MMP-9表达下降有统计学意义(P<0.05)。结论:补肾疏肝方可显著抑制Lewis肺癌生长和自发性肺转移,与顺铂联合有协同作用,其抑制肿瘤生长和转移的机制之一可能为下调VEGF和MMP-9的表达。Objective： To observe the inhibitory effect on growth and metastasis of BuS henS huG an decoction on mice bearing Lewis lung cancer（ LLC） and investigate its underlying mechanism. Methods： Forty C57 BL /6 mice that had been successfully established Lewis lung carcinoma model were randomly assigned to four groups： model group,DDP group,Chinese herbal medicine group and combined group. Model group was given intragastric administration with saline 0. 4 mL everyday; DDP group was given intraperitoneal injection of cisplatin solution 0. 5 mL（ containing DDP 0. 1 mg） on d1,d3,d5; chinese herbal medicine group was given intragastric administration with BuS hen ShuG an decoction 0. 4 mL ·d- 1（ containing raw materials 0. 7 g）; combined group was given intragastric administration with chinese medicine and intraperitoneal injection of cisplatin solution which was the same as Chinese herbal medicine group and DDP group; all the interventions lasted 21 days. All the mice were sacrificed in d22,tumor weight and numbers of spontaneous lung metastasis were measured; the expression of microvessel density（ MVD）,vascular endo-thelia growth factor（ VEGF） protein and matrix metalloproteinase-9 protein（ MMP-9） were detected by immunohistochemistry. Results： Compared with model group,the treatment group could significantly inhibit tumor growth,the difference was significant（ P 0. 01）. Compared with chinese herbal medicine group,the average tumor weight of combined group decreased significantly（ P 0. 01）. Compared with model group,the treatment group could significantly inhibit the spontaneous pulmonary metastasis（ P 0. 05 or P 0. 01）; Compared with DDP group and Chinese herbal medicine group,the number of spontaneous lung metastasis of combined group was significantly Lower（ P 0. 05）. Compared with model group,MVD and VEGF of all treatment groups decreased significantly（ P 0. 05 or P 0. 01）; compared with DDP group,combined decreased significantly（ P 0. 05）. Compared with model
