摘  要:目的:基于NF-κB信号通路观察小青龙汤合玉屏风散对变应性鼻炎大鼠鼻黏膜AQP5含量和AQP5 m RNA表达的影响,揭示小青龙汤合玉屏风散治疗AR的作用机制。方法:将清洁级健康雄性Wistar大鼠48只按体重采用随机区组法分为6组,即分成正常组、AR模型组、Forskolin(c AMP激活剂)干预组、H89(PKA抑制剂)干预组、PDTC(NF-κB抑制剂)干预组、小青龙汤合玉屏风散组,每组8只。采用卵蛋白全身致敏与局部攻击方法制作AR大鼠模型,观察小青龙汤合玉屏风散组治疗后鼻黏膜AQP5含量和AQP5 m RNA表达,结果进行统计学分析。结果:模型组AQP5含量和AQP5 m RNA表达下调,c AMP激活剂Forskolin干预组、NF-κB抑制剂PDTC干预组、小青龙汤合玉屏风散治疗组均能使AQP5含量和AQP5 m RNA表达上调,而PKA抑制剂H89能下调AQP5含量和AQP5 m RNA表达。结论:小青龙汤合玉屏风散治疗变应性鼻炎作用机制可能与抑制NF-κB信号通路和兴奋c AMP-PKA信号通路使AQP5含量和AQP5 m RNA表达上调有关。Objective:To observe the effect of the Xiaoqinglong Decoction and Yupingfeng Power on the content of AQP5 and the expression of AQP5 m RNA in nasal mucosa of allergic rhinitis rats by NF-κB signaling pathway,and reveal the mechanism of the Xiaoqinglong Decoction and YupingfengPower treating AR. Methods:48 healthy male wistar rats were divided into 6 groups by randomized bocks design: normal group,AR model group,Forskolin(c AMP activator) group,H89(PKA inhibitor) group,PDTC(NF-κB inhibitor) group,and Xiaoqinglong Decoction and Yupingfeng Power group,8 rats in each group. The AR rat model was made by the method of whole body sensitization and local attack. The content of AQP5 and the expression of AQP5 m RNA was observed after the treatment of the Xiaoqinglong Decoction and Yupingfeng Power group. The results were statistically analyzed. Results:The content of AQP5 and the expression of AQP5 m RNA in the model group decreased,and the content of AQP5 and the expression of AQP5 m RNA in Forskolin group,PDTC group,XiaoqinglongDecoction and Yupingfeng Power group increased,while the PKA inhibitor H89 could decrease the content of AQP5 and the expression of AQP5 m RNA. Conclusion:The mechanism of the treatment of allergic rhinitis by Xiaoqinglong Decoction and Yupingfeng Power may inhibit the content of AQP5 and the expression of AQP5 m RNA by inhibition of NF-κB signaling pathway and excitatory c AMP-PKA signaling pathway.
