摘  要:目的:研究天麻素对过氧化氢损伤大鼠原代皮层神经元的保护作用。方法:本研究采用过氧化氢损伤大鼠皮层神经元模型,在造模同时给予不同浓度的天麻素溶液,采用DPPH法测定天麻素对氧自由基的清除能力;通过检测尿酸生成,测定天麻素对过氧化氢损伤后大鼠皮层神经元中黄嘌呤氧化酶活性的影响。提取蛋白,通过WB法检测BDNF表达。结果:天麻素能够逆转过氧化氢对大鼠原代皮层神经元造成的损伤,降低细胞上清液中LDH含量,提高活细胞量;细胞上清中SOD活性升高;天麻素可清除氧自由基,抑制黄嘌呤氧化酶活性;天麻素能够促进损伤神经元中BDNF的表达。结论:天麻素对过氧化氢损伤的神经元有显著保护作用。Objective:To explore the protective effect of gastrodine on rat primary cortical neurons after H2O2 injured. Methods: The injured rat primary cortical neuron model was established by H2O2 to examine the antioxidative effect of gastrodine. DPPH method was applied here to measure the free radical scavenging effects of different concentrations of gastrodine. The content of uric acid produced in xanthine/xanthine oxidase system was detected to reflect the effect of gastrodine on xanthine oxidase activity. BDNF expression was measured by WB. Results:Gastrodine protected rat primary cortical neurons from H2O2 injure. LDH in cell culture was reduced and the number of viable cell was increased. The activity of SOD in the neuron culture was increased. Gastrodine could scavenge free radical and inhibit the activity of xanthine oxidase. Gastrodine could induce BDNF expression in the injured neurons. Conclusion:Gastrodine can protect H2O2 injured rat primary cortical neurons.
