摘 要:目的:探究黄柏在一系列的炮制工序中指标性成分的变化情况。方法:采用高效液相色谱法测定采自同一植株的黄柏药材在不同的炮制工序下黄柏碱和小檗碱的含量变化情况。其高效液相色谱的检测条件为Ecosil C_(18)色谱柱(250mm×4.6mm,5μm);流动相:乙腈-0.1%磷酸水(50∶50)(每100mL0.1%磷酸水加十二烷基硫酸钠0.4g);检测波长:284nm;体积流量:1mL/min;柱温:35℃。结果:黄柏在软化和盐炙过程中,其生物碱的含量均有所降低,且软化过程降低得最多;在代表方剂中,盐炙品入药较之生品入药,其检测出的指标成分更多。结论:黄柏饮片加工应该注意炮制过程中的成分流失,并尽可能做到提高黄柏饮片中指标性成分的含量。Objective:To study the changes of index components in a series of processing steps of Phellodendron chinense Schneid. Methods : The contents of berberine, phellodendrine from different processed steps were measured by HPLC from one plant of Phellodendron chinense Schneid. The content determination were carried out on an Ecosil C18 column (250mm×4.6mm, 5μm) with acetonitrile-0.1% phosphoric acid water (50:50) (added 0.4g SDS to 0. 1% phosphoric acid water per 100mL) as mobile phase. The detection wavelength was 284nm and injection volume was 10vtL at the flow rate of 1.0mL/min. The column temperature was set for 35℃. Results: The content of alkaloid was reduced during the processing step of softening and stir frying with salt,but the process of softening can reduce the content more. In the prescription which include the processed product of Phellodendron ehinense Schneid has more alkaloid the the prescription included the raw one. Conclusion : The reducing content of alkaloid in Phellodendron chinense Schneid should be noted. To improve the content of index components in Phellodendron chinense Schneid as far as possible
