摘  要:目的:通过对稳定表达抗汉滩病毒鼠/人嵌合抗体的CHO细胞进行驯化,检测其抗体的表达情况,并对所表达抗体的生物学活性进行初步鉴定。方法:将本课题组前期筛选得到的稳定高表达细胞株1-D9进行无血清驯化,使其适合悬浮培养后接种到摇瓶,通过调整不同的培养条件,每天检测细胞的活率和抗体表达量,并对表达的抗体进行生物学活性鉴定。结果:通过驯化得到了适合悬浮培养的CHO细胞株,其抗体表达量高,生产周期短;并优化了其悬浮培养条件,表达的抗体经鉴定其生物学活性稳定。结论:优化了抗汉滩病毒鼠/人嵌合抗体表达条件,为下一步工业化生产提供了实验依据。Objective: To obtain a modified CHO cell line which can stably express mouse/human chimeric anti-body for anti-hantaanvirus(HTNV) and accommodate to serum-free medium for large scale production. Methods:The modified CHO cell line 1-D9 were adapted to serum-free medium using a gradient reduction of serum man-ner, and were transformed to a suspension cell at a specific condition, which was also adjusted for keeping the an-tibody viability and productivity. Results: We optimized the method of serum-free adaption and the suspended 1-D9. The cell line had advantages over original cell line in both antibody production and growth rate. Conclusion:Our work successfully optimized the condition of chimeric anti-HTNV antibody expression and could be a profit-able trial for industrialization of antibody production.
