摘 要:目的:探讨免疫刺激剂脂多糖对镉处理后的河南华溪蟹是否有调节作用,以及脂多糖的作用机理。方法:设置5个脂多糖处理组(0.5、1.0、2.0、4.0和8.0μg/m L)和1个对照组(无菌生理盐水),分别刺激溪蟹12、24和48 h后,根据机体中血淋巴细胞总数和溶菌酶活力的变化,选取脂多糖最佳作用条件;镉暴露溪蟹96 h后,按最佳作用条件注射脂多糖(8.0μg/m L,24 h),测定血清中溶菌酶和酸性磷酸酶活力。结果:脂多糖刺激溪蟹后,血细胞数量和溶菌酶活力的变化表现出时间-剂量效应;镉暴露后注射脂多糖,溪蟹血清中溶菌酶活力无显著变化,而酸性磷酸酶活力与镉单独作用组相比显著升高。结论:脂多糖对河南华溪蟹免疫系统有良好的刺激作用,该研究为探讨脂多糖对甲壳动物刺激作用机理提供了理论基础,同时也可为脂多糖作为免疫增强剂在水产养殖中的研究应用提供参考。Objective: To observe the effects of immunostimulant lipopolysaccharide(LPS) on freshwater crab Sinopotamon henanense exposing to cdmium, and it's mechanism of action. Methods: A series of LPS concentrations(0.5, 1.0, 2.0, 4.0 and 8.0 μg/m L) and a control group were designed to study the stimulating effect on thecrabs. The optimal action condition was chosen, according to the changes of total hemocytes number and activityof lysozyme in hemolymph of crabs after injecting with LPS for 12, 24 and 48 h. Then, LPS was injected afterthe organisms exposed to cadmium, and the activities of lysozyme and acid phosphatase were determined. Results:The changes of total hemocytes number and activity of lysozyme in crab showed time-dose dependent effect afterLPS injection. The activity of lysosome showed no statistically changes in crabs with cadmium esposure followedby LPS stimulation. However, compared with the cadmium treatment group, the activity of acid phosphatase wasup-regulated after crab were injected with LPS. Conclusion: LPS displayed a good stimulating effect on the im-mune system of the crab S.henanense. This study provided a theoretical basis for the stimulation mechanism ofLPS, and also provided a reference value for the research and application of LPS as an immune enhancer in aqua-culture.
