摘  要:目的探究DNA低甲基化状态相关的基因在系统性红斑狼疮患者外周血单核细胞(PBMC)中的表达水平。方法选取湘雅三医院于2015年1-12月收治的SLE患者40例,分为缓解期组(SLEDAI<5)与活动期组(SLEDAI≥5),每组各20人。同时选取健康志愿者20例作为对照组。测定其PBMC中mbd2、CD11a基因表达水平,分析结果并探究其表达水平对DNA低甲基化状态的影响。结果缓解期mbd2、CD11a基因表达水平分别为(0.210±0.040、0.812±0.214);活动期组mbd2、CD11a基因表达水平分别为(0.368±0.055、1.214±0.374)。缓解期组与活动期组mbd2、CD11a基因表达水平明显高于对照组(0.061±0.013、0.413±0.103),活动期组CD11a基因表达水平明显高于缓解期组,差异有统计学意义(P<0.05)。结论 mbd2、CD11a基因在患者PBMC中存在过度表达,并且与患者发病状况关系密切。Objective To explore the expression level of DNA in low methylation status in peripheral blood mononuelear cells ( PBMC ) of patients with systemic lupus erythematosus ( SLE ). Methods Forty patients with SLE hospitalized in the Third Xiangya Hospital from January to December 2015 were selected to divide into the remission phase group (SLEDAI〈5 ) and the active phase group ( SLEDAI ≥ 5 ), with each n = 20. At the same time, 20 healthy volunteers were selected as the control group. We determined the expression levels of mbd2 and CD11a in PBMC, analyzed the results and explored the effect of their expression levels on DNA in low methylation status. Results The expression levels of mbd2 and CDlla genes in the remission phase group ( (0.21±0.040), (0.812±0.214)) and the active phase group ( (0.368±0.055), (1.214±0.374) ) were all significantly higher than those of the control group ( (0.061±0.013), (0.413±0.103) ). The expression level of CDlla gene in the aetive phase group was significantly higher than that in the remission phase group, with a statistieally significant difference ( P〈0.05 ). Conclusions The mbd2 and CDlla genes overexpress in PBMC of SLE patients, moreover, they are closely related to the ineidenee of the patients.
