乌丹降脂胶囊对NAFLD大鼠肝脏NF-κB表达及血清生化指标的影响

摘　　要：目的研究乌丹降脂胶囊对非酒精性脂肪肝(NAFLD)大鼠肝脏核因子-κB(NF-κB)表达及血清生化指标的影响。方法将40只雄性SD大鼠随机分为正常组、模型组、易善复组和乌丹降脂胶囊组,每组10只。正常组以基础饲料喂食,其余3组以高脂饲料喂食,2周后,正常组和模型组给予生理盐水10 mL/(kg·d)灌胃,易善复组以浓度为8.523 g/L的易善复溶液10 mL/(kg·d)灌胃,乌丹降脂胶囊组以浓度为41.667 g/L的乌丹降脂胶囊溶液10 mL/(kg·d)灌胃,均连续14周。检测各组外周血ALT、AST、TC、TG、HDL-C、LDL-C、C反应蛋白(CRP)、胰岛素(Insulin)和肿瘤坏死因子-α(TNF-α)水平及肝脏组织中NF-κB p65蛋白表达量。结果模型组HDL-C水平明显低于正常组(P<0.05),其余各指标水平均明显高于正常组(P均<0.05);易善复组与乌丹降脂胶囊组CRP、LDL-C水平均明显高于正常组(P均<0.05)。易善复组与乌丹降脂胶囊组CRP、ALT、AST、TC、TG、LDL-C、Insulin、NF-κB水平均明显低于模型组(P均<0.05),HDL-C水平明显高于模型组(P均<0.05),乌丹降脂胶囊组TNF-α水平明显低于模型组(P<0.05);乌丹降脂胶囊组CRP、LDL-C、TNF-α水平均明显低于易善复组(P均<0.05)。结论乌丹降脂胶囊可有效降酶降脂,减轻炎症,促进肝细胞功能恢复,其抑制肝脏NF-κB的表达可能是治疗NAFLD的机制之一。Objective It is to study the effect of Wudan lipid-lowering capsule on the expression of NF-κB in liver and ser- um biochemical indicators in the rats with NAFLD. Methods Forty male SD rats were randomly divided into four groups： con- trol group, model group, Essentiale group and Wudan lipid-lowering capsule group, 10 rats in each group. The control group was fed with basic food, the remaining three groups were fed with high fat food. After two weeks, the control group and model group were given normal saline 10 mL/（kg· d） by lavage, Essentiale group was given 10 mL/（ kg · d） Essentiale complex solution whose content was 8. 523 g/L by lavage,Wudan lipid-lowering capsule group was given 10 mL/（ kg · d） Wudan lipid- lowering capsule complex solution whose content was 41. 667 g/L by lavage. The treatment courses were 14 weeks. The con- tent of peripheral blood ALT, AST, TC, TG, HDL-C, LDL-C, CRP, insulin, and TNF alpha and the expression of NF-κB p65 protein in liver tissue were detected. Results Compared with control group, the levels of HDL-C were lower while the oth- er indexes were higher in model group（P 〈 0. 05）. The levels of CRP and LDL-C in Essentiale group and Wudan lipid-lowering capsule group were increased significantly compared with control group, the differences were statistically significant （P 〈 0.05）. Compared with model group, the levels of CRP, ALT, AST, TC, TG, LDL-C, insulin, NF-κB in Essentiale group and Wudan lipid-lowering capsule group were decreased significantly, the levels of HDL-C were increased significantly, the level of TNF alpha in Wudan lipid-lowering capsule group was obviously decreased, the differences were statistically significant （P 〈 0.05）. Compared with Essentiale group, the levels of CRP, LDL-C,TNF alpha of Wudan lipid-lowering capsule group were decreased significantly,the difference was statistically significant（P 〈 0.05）.
