摘 要:目的:评价关黄柏的质量以及其总生物碱的含量。方法:酸性染料比色法,在pH=5.2条件下,生物碱与酸性染料结合,即生成有吸收光谱明显红移的有色离子对,该离子对可以定量的被有机溶剂萃取,测定有机相中有色离子对特征波长处的吸光度,从而再进行关黄柏中总生物碱含量的测定,并进行回收率实验。结果:在415 nm处有最大吸收,酸性染料比色法的显色条件为pH=5.2的缓冲溶液8.0 mL,溴麝香草酚蓝溶液3.0 mL,氯仿萃取三次,每次4.0 mL,每次萃取振摇3分钟,静置0.5 h。盐酸小檗碱在55.2~166.5μg/mL范围内成线性关系,平均回收率为100.5%,RSD为2.0%(n=5)。结论:该方法用量少,灵敏度高、重复性好。具有一定的专属性和准确度。Subjective: Content quality evaluation of Cortex Phellodendri and its total alkaloids. Berberine hydrochloride as in 55.2~166.5 μg/mL range linear relationship, the average recovery was 100.5 %, RSD was 2.0 %(n=5).
