摘 要:为建立小鼠化学性、药物性肝损伤模型及大鼠腓肠肌损伤模型,给予长梗冬青苷(PE)及辛伐他汀(SIM)干预,分别检测小鼠血清谷草转氨酶(AST)、T淋巴细胞白血病(ALT)水平和大鼠血清肌酸激酶(CK)、肌酸激酶同工酶(MB)水平。制备小鼠肝脏、大鼠腓肠肌组织切片,光镜观察组织形态学变化,分析PE对肝损伤、肌损伤有无影响。结果表明:PE对肝损伤小鼠血清AST、ALT水平升高、肌损伤大鼠血清CK、MB水平上升均呈抑制作用;组织形态学观察显示PE可改善肝损伤小鼠肝纤维化,抑制肝脏炎性细胞浸润。PE对肌损伤大鼠肌纤维断裂、肌间质肿胀有显著改善作用。说明PE对化学性与药物性肝损伤及物理性肌损伤无不良影响且有改善作用;PE对SIM加重的肝损伤与肌损伤亦有一定的改善作用。To explore the effects of pedunculoside(PE)on the chemical and drug-induced liver injury in mice and the gastrocnemius damage in rat.The models mentioned above were gavaged with PE or simvastatin(SIM)to investigate the effect of PE on mice and rat.We detected the serum levels of AST,ALT in mice and the contents of CK,MB in rat,and observed the pathological changes of liver tissue in mice with hepatotoxicity.The results showed that PE could reduce the level of AST and ALT in mice with liver injury,as well as the contents of CK and MB in rat of gastrocnemius injury.Moreover,PE effectively improved the degeneration of liver cell,and reversed the inflammatory cells infiltration,which contributed to the improvement of hepatic pathological morphology in mice with hepatotoxicity.Additionally,PE could improve both myofibril fragmentation and interstitial muscle swelling.Meanwhile,PE could reduce the inflammatory cells infiltration in rat.It was concluded that PE had protective effect on chemical or drug-induced liver injury in mice and gastrocnemius damage in rat.In addition,PE could reverse the damage accelerated by SIM.
