摘 要:目的 确定常染色体STR分型在叔侄关系排查时的全不同基因座预测界值,探讨叔侄关系在案件排查尤其是在Y-STR DNA数据库比对合成技战法中的应用策略。方法 以已知叔侄对作为实验组,无关个体以及同一祖先23~27个Y-STR等位基因相同但超出五代亲缘关系的个体作为对照组;分别采用ID Plus及PP21系统进行常染色体STR检测。结果 叔侄对全不同基因座及共有等位基因数分布情况与对照组存在显著性差异;同一祖先23~27个Y-STR等位基因相同的个体对于叔侄关系的判断有一定的影响。基于两个体23~27个Y-STR等位基因相同的前提,若检测ID系统15个常染色体STR基因座时其全不同基因座≤4或检测PP21系统20个常染色体基因座时全不同基因座≤5,则提示两个体可能有叔侄关系。结论 在应用Y-STR DNA数据库比对合成技战法进行家系排查时,结合常染色体STR分型进行叔侄关系预测以寻找嫌疑人的方法具有可行性,以全不同基因座数目预测叔侄关系的方法简便高效。Objective To define the predictive threshold-value of the entirely different loci of autosomal STR for deciding the paternal uncle-nephew relationship and discuss the role of such kinship in case Inoestigation, especially in the comprehensive utilization ofY-STR DNA database. Methods With the paternal uncle-nephew pairs as the experimental group, the control group is consisted of the irrelative individuals and those descending beyond 5 generations from the same ancestor yet having the identical 23-27 Y-STR allelic genes. The ID Plus and PP21 systems are applied to test the above individuals' autosomal STR genotypes. Results There are obvious differences in the distribution of both the entirely different loci and the identical allele number between the two groups; the individuals, descending beyond 5 generations from the same ancestor but owning the identical 23-27 Y-STR allelic genes, can certainly influence the judgment of the paternal uncle-nephew relationship. On the prerequisite of same 23-27 Y-STR allelic genes commonly owned by the two individuals, the predictive threshold value of entirely different loci is no greater than 4 for case Inoestigation when the 15 loci of the ID system are tested. Meanwhile, such value should be no larger than 5 out of the 20 loci in the PP21 system. Both of the above predictions imply the possibility of the paternal uncle-nephew relationship. Conclusion It is feasible to target the criminal suspect in case Inoestigation by combination of the comprehensive utilization of Y-STR DNA database with autosomal STR genotyping. The way to predict the paternal uncle-nephew relationship is facilitative, convenient and efficient by the number of the entirely different loci.
