角类肥蛛体内共生菌多样性检测

摘  要:本研究以角类肥蛛(Larinioides cornuta)为实验材料,采用DNA提取试剂盒提取角类肥蛛的基因组DNA,把10头角类肥蛛的基因组DNA按照等摩尔量混合,以混合DNA为模版,用细菌16SrDNA的1对通用引物27F和1487R进行PCR检测并进行序列测定。检测结果:角类肥蛛只感染了Spiraplasma1个属的内共生菌,50个克隆测序结果共获得11个不同的Spiraplasma序列(KX559370.KX559380)。所获得的序列与NCBI核苷酸库的比对结果显示,角类肥蛛体内检测到的Spiraplasma与鞘翅目的1种象鼻虫( Curculio elephas )和瓢甲科Anisosticta属昆虫体内检测到的Spiraplasma相似度较高(98%-99%);所获序列与NCBI核糖体RNA数据库的比对结果显示,角类肥蛛体内的Spiraplasma共生菌与Spiroplasmaixodetis相似度最高(98%-99%)。In this study, L. cornuta was used for the extraction of genome DNA. 10 individual DNA of L. cornuta were blended according to the same DNA molar quality. A target fragment was amplified with the template of blended DNA, and the primer was 27F and 1487R of 16S r DNA. The results showed that only Spiroplasma was detected in L. cornuta, and 11 sequences(KX559370-KX559380) of Spiroplasma were obtained. The result of blast with NCBI nucleotide collection showed the Spiroplasma from L. cornuta was similar to the Spiroplasma from Curculio elephas and the insects of genus Anisosticta (98%-99%). The result of blast with NCBI 16S ribosomal RNA of GenBank showed Spiroplasma obtained in this study was similar to Spiroplasma ixodetis(97%-98%).
