摘 要:为了研究蝴蝶兰SOC1基因的功能,为蝴蝶兰分子育种等方面的研究提供一定的理论依据。本研究以蝴蝶兰"聚宝"盛花期的花瓣为材料提取总RNA,采用RT-PCR方法克隆蝴蝶兰SOC1基因的编码区序列,片段长度为682 bp,共编码219个氨基酸,该基因命名为SOC1,登录号为:KT852838。对蝴蝶兰SOC1基因进行同源分析及构建系统发育树,分析结果显示,与SOC1核苷酸序列同源性最高的是小兰屿蝴蝶兰(Phalaenopsis equestris),达到93.72%,SOC1基因与兰科植物小兰屿蝴蝶兰及石斛的亲缘关系最近。实时荧光定量PCR分析表明,SOC1基因在的不同时期不同部位都有表达,但表达丰度不一致。在根中,营养生长期表达量最高;在叶中,抽葶期表达水平最高;在花葶中,抽葶期、开花期和子房发育期的表达水平相当;在花器官中,以蕊柱的表达量最高,其次是子房,而花萼、唇瓣、侧瓣中只有微量表达。研究认为SOC1基因在蝴蝶兰生长发育中既调控营养生长,又调控生殖生长;在花器官中主要参与蕊柱和子房的发育。In order to study the function of SOC1 gene in Phalaenopsis,and to provide a theoretical basis for the study on molecular breeding and other aspects of Phalaenopsis.Total RNA was extracted from the flower of Phalaenopsis'Jiuhbao'in the stage of flowering.The conserved sequence of SOC1(682 bp,219 amino acids) gene has cloned by using the RT-PCR technology.The sequence was registered in Gen Bank with accession number KT852838.Multiple sequence alignments and phylogenetic tree analysis showed 93.72% homologies with Phalaenopsis equestris.The result of analysis presented that SOC1 gene was the closest relatives with Phalaenopsis equestris and Dendrobium.SOC1 gene was analyzed by real-time fluorescence quantitative PCR,which showed that it was expressed in different position at different times,whereas the expression of abundance is different.In root,the expression level of vegetative growth stage was the highest.In leaf,the expression level of scape forming stage was the highest.In the scape,the expression level in the reproductive growth stage,in flowering stage,and the ovary development stage were commendable,and the difference of them was insignificant.In the floral organs,the expression level of gynostemium was the highest,followed by the ovary,and were very little in the calyx,labellum and side-lobe.We speculated that SOC1 gene not only regulates the vegetative growth and reproductive growth in Phalaenopsis development,but also mainly participates in the growth of gynostemium and ovary in floral organs.
