摘  要:PFT1/MED25(PHYTOCHROME AND FLOWERING TIME1/MEDIATOR25)在植物中组织发育、成花诱导及胁迫响应等多种进程中起重要调控作用。本研究采用RT-PCR和RACE技术结合的技术克隆了红掌PFT1/MED25基因(Aa PFT1)全长2 457 bp的c DNA序列,推测其含有1个1 974 bp的完整开放阅读框(ORF),编码658个氨基酸。定量RT-PCR分析表明:Aa PFT1在红掌‘热情’各组织中都有表达,其中根的表达量较高;苗期表达量高于成年期;在叶和花的不同组织发育阶段,Aa PFT1在幼嫩组织和成熟组织中表达量较高。Aa PFT1在一年的生长周期内,表现出动态表达变化,但在叶片、苞片、花序中的变化趋势不同。红掌‘卫城’的外源激素处理表明:Aa PFT1能被SA诱导迅速诱导上调,而其他激素处理早期无显著变化。这些结果为进一步解析Aa PFT1基因在红掌中可能的多重作用机制奠定基础。PFT1/MED25(PHYTOCHROME AND FLOWERING TIME1/Mediator subunit 25) plays important roles in controling plant tissues development,floral induction and stress response.In this study,Anthurium(Anthurium andraeanum Linden) PFT1/MED25-like gene(Aa PFT1/MED25) was cloned by reverse transcription PCR and rapid amplification of c DNA ends,which has 2457 bp and is speculated the full-length c DNA containing a 1 974 bp ORF encoding a putative protein with 658 amino acids.Quantitative RT-PCR analysis indicated that Aa PFT1 of Anthurium andraeanum.CV.‘Tropical'is ubiquitously expressed in various tissues and relatively predominant in roots and the expression at adult stage was higher than that at the seedling stage.It is also abundant in young and mature tissues during development stages.Monthly change dynamics of Aa PFT1 expression patterns are distinguished from leaf,spath and spadix.The result of exogenous hormone treatments showed that Aa PFT1 of Anthurium andraeanum.CV.‘Acropolisl'could be rapidly induced by the artificial treatment of SA,while the other phytohormone had no significant change in early period.All these results will facilitate to expand our understanding about the multiple regulations involved by Aa PFT1 in Anthurium.
