摘  要:三色堇花色艳丽,品种丰富,是重要的冬春季花卉。本研究采用黄底紫斑品种‘梦蝶’为研究材料,克隆花瓣中可能参与花青素累积起调控作用的MYB基因VwMYB8,结果显示:克隆得到的cDNA全长为1 078 bp,经ORF Finder分析发现其有长度为849 bp的开放阅读框,编码282个氨基酸;构建瞬时表达载体pXCS-VwMYB8,采用粒子轰击的方法作用于月季‘粉扇’花瓣细胞,经过48 h培养后观察转化材料细胞中色素的变化,转化的细胞颜色明显加深,表明基因VwMYB8可以提高月季花瓣细胞中色素的含量。Viola ×wittriki anan is an important flower in winter and spring for its beautiful flower colors and rich cultivars. In this study the yellow cultivar‘Mengdie'with purple blotch on petals was used as research material to clone VwMYB8 of MYB gene in the petal which may be involved in regulation of anthocyanin accumulation. The results showed that the c DNA full length of this gene was 1 078 bp. ORF Finder analyses showed that the open reading frame was 849 bp, encoding 282 amino acids. A transient expressed vector p XCS-VwMYB8 was constructed, using particle bombardment assay to transfer this gene into the petal cell of rose‘Pink fan'. The distinct pigmentation was observed in the transformed rose petal cells after 48 h incubation. The result of transient expression indicated that VwMYB8 gene may improve pigment accumulation in cells of rose petals.
