摘 要:利用MISA软件对短丝木犀花和叶芽转录组测序得到的92 798条独立基因(unigenes)进行SSR位点查找和分析,共搜索到11 881个SSR位点,除单碱基重复外,SSR出现频率为4.64%,分布密度为1/15.02 kb。二碱基和三碱基重复为短丝木犀SSR主要重复单元类型,分布占SSR总数的66.35%和30.42%。其中AG/CT和AAG/CTT分别为二碱基和三碱基重复单元的优势重复基元,分别占各自重复单元的63.21%、30.38%。短丝木犀SSR多为重复长度小于20 bp的短序列,长度大于20 bp的SSR仅占总数的13.47%。同时发现短丝木犀SSR的频率和长度呈显著负相关(p<0.01),相关系数为-0.408。另外,设计筛选得到2 435对SSR引物。短丝木犀转录组SSR位点出现频率高,类型丰富、多态性潜能较高。Based on the Illu mina hi gh throughput sequencing data of flowers and leaf buds of Osmanthus serrulatus, 10 497 unigenes with 11 881 SSR loci were detected from all of 92 798 unigenes with MISA software.In the 2-6 bases repeat microsatellites, a total of 4 306 SSRs were identified in 4 027 unigenes, with an occurrence frequency of 4.64% and an average density of one SSR per 15.02 kb. Dinucleotide and trinucleotide repeat microsatellites were the main SSR types of O. serrulatus, accounting for 66.35% and 30.42% of all SSRs,respectively. Among all the 61 SSR motifs, AG/CT was the most abundant motif(1806, 63.21%) in dinucleotide repeats, and AAG/CTT was most(398, 30.38%) in trinucleotide repeats. The SSR in length below 20 bp were in maximum proportion, while the SSR over 20 bp were only 13.47%. There were significant negative correlation(p<0.01) between the frequency of SSR and the length, with correlation coefficient of-0.408. Additionally, a total of 2 435 primer pairs were designed for marker development.
