Construction of Encryption Map and Mapping QTLs For Fruit-Associated Traits in Cucumber(Cucumis sativus L.)

摘  要:本研究以黄瓜纯系S94(华北类型)和S06(欧洲类型)为亲本,通过单粒传法获得包含224个株系的F6:7代重组自交系(RIL)群体。结合分子遗传加密图谱的构建及不同季节(F7/秋,F7/春两季)黄瓜重要果实性状表型调查数据,采用Win QTLcart 2.5软件复合区间作图法,对9个黄瓜果实相关性状(瓜长,瓜重,瓜把长,瓜横径,果柄长,果肉厚度,心腔直径,及长把比和长径比)QTL位点进行检测;同时,对果实质量性状无光泽果皮(D)、一致果皮颜色(u)和小刺基因(ss)进行了定位。结果显示:(1)构建包含7个连锁群,610个标记的遗传图谱,覆盖基因组749.2 c M,平均标记间距为1.2 cM;(2)果实相关的9个性状在F7/秋与F7/春两季共检测73个QTLs位点(F7/秋52个,F7/春49个),分布于全部7条染色体上,其中在果实相关性状中,仅瓜重fw4.1在春秋两季稳定检测到且贡献率较高,春季14.7%和秋季21.7%,其余位点高的贡献率(>10%)都在单一季节表现;(3)黄瓜果实无光泽果皮(D)、一致果皮颜色(u)和小刺基因(ss)定位于第5染色体。We used the cucumber pure lines S94(Northern China type) and S06(Northern European type) as the parent, and by using single seed descend method to obtain F6:7generation recombinant inbred line(RIL) population which contains 224 lines. We based on the construction of molecular genetic encryption map and the investigation data of important fruit characters of cucumber in different seasons(F7/autumn, F7/spring and two seasons), and we used the winqtlcart 2.5 software composite interval mapping method to test 9 cucumber fruit-related traits QTL loci(fruit length, fruit weight, fruit stalk length, fruit diameter, fruit pedicel length, fruit 覱esh thickness, seed-cavity diameter, fruit length/stalk ratio and fruit length/diameter ratio). At the same time, we position the dull fruit skin(D) uniform immature fruit color(u) and small spines(ss). The result showed that(1) we constructed a genetic map with 7 linkage groups, 610 markers, covering 749.2 c M, with an average marker density of 1.2 c M;(2) 9 traits related to fruit both in F7/ autumn and F7/ spring season were detected 73 loci(F7/ autumn 52, F7/ spring 49) and these distributed in all seven linkage groups, which in fruit traits, only fruit weight fw4.1 in stable detection of spring and autumn and contribution rate is higher, the spring 14.7% and the autumn 21.7%, with the rest of the site high rate(>10%) in a single season;(3) Dull fruit skin(D), uniform immature fruit color(u), small spines(ss) located on chromosome 5th.
