摘  要:目的:建立硫糖铝中铝离子(Al^(3+))在大鼠胃组织残留量的测定方法,考察硫糖铝对胃溃疡粘膜的靶向粘附作用。方法:采用干法灰化消解胃组织样品,电感耦合等离子体-原子发射光谱法(ICP-AES)测定给药后不同时间点大鼠胃组织中Al^(3+)的残留量。ICP-AES检测波长为396.153 nm,检测器为电感耦合阵列检测器,载气为氩气。结果:干法灰化大鼠胃溃疡组织,样品消解完全,可满足ICP-AES的检测要求;在选定的最佳条件下用ICP-AES检测Al^(3+)的检出限为8 ng·m L^(-1),样品间Al^(3+)残留量相对偏差小于13.4%,回收率在96.0%~99.5%之间;给药0.5~4 h时间段内,溃疡胃组织中的Al^(3+)残留率为正常胃组织的2倍以上;给药6 h后,正常胃组织中Al^(3+)残留率显著下降,仅为0.84%,而溃疡胃组织在给药16 h后仍高达29%。结论:ICP-AES测定大鼠胃组织中Al^(3+)的残留量准确可行;硫糖铝对胃溃疡粘膜的粘附性显著高于正常胃粘膜,具有良好的靶向性。Objective: To study the targeted adhesion for sucralfate to impaired gastric mucosa of SD rats by determining the residual of aluminum ion (Al3+)in the mucous.Methods: The stomach tissue samples were digested by dry-ashing method.The residues of Al3+ in rat stomach tissues at different time points were determined by inductively coupled plasma atomic emission spectrometry (ICP-AES)at the wavelength of 396.153 nm.The detector was charge-coupled device array detector (CCD), and the carrier gas was argon.Results: The stomach tissue samples digested by dry-ashing method could meet the request for ICP-AES detection.In the optimal conditions, the detection limit of Al3+ was as low as 8 ng·mL-1, and the relative standard deviation (RSD)of Al3+ content was less than 13.4%, which satisfied the request of biological specimen analysis.The obtained recovery values were in the range from 96.0% to 99.5%.Between 0.5 h and 4 h after the intragastric administration of sucralfate, the residue level of Al3+ in the impaired gastric was more than 2 times as the normal tissue.6 h after the administration, the residual percent of Al3+ in normal mucosa significantly descended, only 0.84%, but that of impaired gastric mucosa was about 29% even 16h after the administration.Conclusion: The determination of Al3+ residual in rat gastric tissue by ICP-AES was accurate and feasible.The adhesion of sucralfate to gastric ulcer mucosa was significantly higher than that of normal gastric mucosa, which indicated that the sucralfate had a selective affinity to the impaired gastric mucosa.
