摘 要:目的:建立同时测定艾叶中6个有机酸类成分含量的一测多评法。方法:采用Thermo Syncronis C18色谱柱(4.6 mm×250 mm,5μm),流动相为0.1%甲酸水溶液-乙腈,梯度洗脱,流速0.8 m L·min^(-1),检测波长325 nm,柱温30℃。以绿原酸为内参物,分别计算与新绿原酸、隐绿原酸、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸的相对校正因子,实现一测多评。同时利用外标法和一测多评法测定6个有机酸类成分的含量。比较2种方法的测定结果是否存在差异。结果:一测多评法与外标法测定结果无显著差异。实验所测得的相对校正因子重复性良好。6批不同产地艾叶样品中新绿原酸、绿原酸、隐绿原酸、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸的含量范围分别为0.15~0.58、0.50~2.65、0.27~0.89、0.62~4.29、1.18~10.72、0.99~6.69 mg·g^(-1)。结论:一测多评法可以用于艾叶中6个有机酸类成分的含量测定,且具有可行性与经济性。Objective: To establish a quality evaluation method to simultaneously determine the contents of six organic acids in Artemisiae Argyi Folium with a single-marker (QAMS).Methods: An HPLC method was established with a Thermo Syncronis C18 (4.6 mm×250 mm, 5 μm) with gradient elution using 0.1% formic acid solution-acetonitrile as the mobile phase.The flow rate was 0.8 mL·min-1, the detection wavelength was 325 nm, and the column temperature was 30℃.Chlorogenic acid was used as the internal reference substance.The relative correlation favtors (RCF) of neochlorogenic acid, cryptochlorogenic acid, 3, 4-dicaffeoylquinic acid, 3, 5-dicaffeoylquinic acid, 4, 5-dicaffeoylquinic acid to chlorogenic acid were calculated.The content of these six organic acids were determined by the external standard method and QAMS method respectively.The QAMS method was validated through comparison of the results obtained by the two different methods.Results: No significant difference was found between the two methods.The RCF showed good reproducibility.The contents of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, 3, 4-dicaffeoylquinic acid, 3, 5-dicaffeoylquinic acid and 4, 5-dicaffeoylquinic acid in 6 batthes of samples of Artemisiae Argyi Folium were 0.15-0.58 mg·g-1, 0.50-2.65 mg·g-1, 0.27-0.89 mg·g-1, 0.62-4.29 mg·g-1, 1.18-10.72 mg·g-1 and 0.99-6.69 mg·g-1, respectively.Conclusion: The QAMS method showed favorable feasibility and economy, which can be used to determine the content of six organic acids in Artemisiae Argyi Folium.
