藏药二十五味珍珠丸中肉桂酸的含量测定及HPLC指纹图谱研究

摘　　要：目的:建立藏药二十五味珍珠丸的HPLC指纹图谱分析方法及肉桂酸含量测定方法。方法:采用ZORBAX SB-C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-0.1%磷酸溶液为流动相,流速1.0 m L·min^(-1),检测波长285 nm,柱温30℃。结果:肉桂酸进样量在0.156~1.404μg的范围内有良好的线性关系(r=0.999 9),平均回收率(n=9)为99.19%。19批二十五味珍珠丸中肉桂酸的含量在30.13~161.08μg·g^(-1);指纹图谱中检出15个共有峰,相似度计算结果在0.887~0.985之间,并通过聚类分析,将不同企业的19批样品分为两类。结论:该方法较好地反映了二十五味珍珠丸的化学成分信息,且操作简单,重复性好,为进行二十五味珍珠丸的质量评价提供了参考。Objective： To establish a method for the determination of cinnamic acid and the analysis of the HPLC fingerprint of Ershiwuwei Zhenzhu pills.Methods： A ZORBAX SB-C18 （4.6 mm×250 mm, 5 μm）column was used with acetonitrile-0.1% phosphoric acid solution at a flow rate of 1.0 mL·min-1 as the mobile phase. The detection wavelength was 285 nm and the column temperature was 30.Results： The method showed good linear relationship within the range of 0.156-1.404 μg for cinnamic acid （r=0.999 9）, and the average recovery （n=9） was 99.19%. For 19 batches of samples, the cinnamic acid content ranged from 30.13 to 161.08 μg·g-1. Fifteen common peaks were selected as characteristic peaks in the chromatogram of Ershiwuwei Zhenzhu pills, and the similarities of chromatographic fingerprint ranged from 0.887 to 0.985. The samples from different companies were classified into two groups based on principal component cluster analysis. Conclusion： The method can well display the chemical information of Ershiwuwei Zhenzhu pills.The method is simple and reliable, and can be used for the quality control of Ershiwuwei Zhenzhu pills.
