摘  要:目的:比较3种中药方剂对亚慢性氟中毒大鼠脂质过氧化物的清除能力,探讨中药复方制剂治疗慢性氟中毒的机制。方法:224只SD大鼠分为正常对照组、中氟组、高氟组和高氟低蛋白低钙组,后3组大鼠给予相应含氟饲料或低蛋白低钙饲料喂养制作亚慢性氟中毒大鼠模型;造模180后,中氟组、高氟组及高氟低蛋白低钙组大鼠又分为模型组、甲方剂组、乙方剂组和丙方剂组,后3组大鼠给于相应中药方剂治疗90 d;观察各组大鼠实验过程中的体重增长、氟斑牙情况,并与中药治疗90 d时取肝脏和肾脏制作组织匀浆检测过氧化氢酶(CAT)活性和丙二醛(MDA)含量。结果:各模型组组织匀浆CAT含量均高于正常对照组(P<0.05);在给予丙方剂治疗后,各个治疗系列中肝/肾组织匀浆CAT含量都有所降低(P<0.05),说明丙方剂对降低CAT水平有作用,即能提高脂质过氧化物的清除能力;各模型组肾脏组织匀浆MDA含量高于正常对照组(P<0.05);给予甲方剂和乙方剂治疗后,肝/肾组织匀浆MDA含量有所降低,说明甲方剂和乙方剂对降低MDA水平有作用,即能提高脂质过氧化物的清除能力,其中乙方剂较甲方剂效果更好。结论:丙方剂能调节CAT含量的能力,使其能恢复到清除H2O2的正常范围;甲方剂和乙方剂可抑制MDA含量升高,其中乙方剂最好。Objective: To explore the effect of three Chinese medicine formulae which may enhance ability of eliminating lipid peroxide of chronic fluorosis rats, and discuss relevant mechanism. Methods: 224 rats were divided into 4 groups: control group, medium-dose fluorine group, high-dose fluorine group, high-dose fluorine supplemented with low protein and low calcium diet group. The lat- ter three groups were fed with fluorine containing feed or low protein low calcium feed, in order to pro- duce sub chronic fluorosis rats; after model producing for 180 days, the latter three groups were further divided into model group, prescription A group, prescription B group and prescription C group. These three new groups were treated with traditional Chinese Medicine formula for 90 days; observing weight gain and dental fluorosis condition, and make tissue homogenate to test CAT activity and MDA content with liver and kidney after 90days of treatment. Results: Compared with control group, the CAT of positive control group was increased which was statistically significant ( P 〈 0.05 ). After given treat- ment of prescription C, CAT in both liver and kidney had been decreased implying prescription C is ef- fective for decreasing the level of CAT, which enhanced ability of eliminating lipids peroxide. Com- pared with control group, the MDA content of model groups were higher than that of control group(P 〈 0.05). After given treatment of prescription A and B, MDA in both liver and kidney had been de- creased implying prescription A and B are effective for decreasing the level of MDA, which enhanced ability of eliminating lipids peroxide, meanwhile prescription B is more effective than prescription A. Conclusion: Prescriptions C are able to adjust the content of CAT, and make it back to the normal range to eliminate H202. Prescription A and prescription B are able to restrain the content of MDA, prescription B is more effective.
