非洛地平导致的外周性水肿与中国人群中CACNA1C基因多态性的相关性分析

摘　　要：目的:探索中国人群中CACNA1C基因单核苷酸多态性(SNPs)与非洛地平导致的外周性水肿是否存在相关关系。方法:纳入190例服用非洛地平片的高血压病患者,分为非水肿组和水肿组2组。用PCR直接测序法检测CACNA1C基因单核苷酸多态性(SNPs),并对rs1051375、rs2238032、rs2239050和rs2239128等位点进行基因分型。用χ2检验方法分析SNPs位点各基因型在2组中的差异。结果:2组中rs1051375、rs2238032、rs2239050的各基因型的频率分布无统计学意义(P>0.05),而rs2239128TT基因型在水肿组分布明显高于非水肿组(P<0.05)。结论:CACNA1C基因的rs2239128TT基因型与非洛地平导致的外周性水肿有明显的相关关系。Objective：To evaluated the association of calcium channel voltage-dependent L type,alpha 1C subunit gene（CACNA1C）pol-ymorphism with the effectiveness of Peripheral oedema adverse drug reaction of Felodipine.Methods：190 cases of hypertensive patients were selected and given Felodipine.They were divided into peripheral oedema group and no peripheral oedema group.The distribution of CACNA1C polymorphism 190 patients was investigated via the polymeras chain reaction and bidirectional sequencing.The general clinical features and polymorphism of CACNA1C gene（rs1051375,rs2238032,rs2239050,rs2239128 allelic loci）were compared among 2 groups. Corralation between CACNA1C gene single nucleotide polymorphism and peripheral oedema of Felodipine for hypertension were analyzed with Chi square test.Results：The CACNA1C genotype of rs1051375,rs2238032,rs2239050 distributions were.not statistically signficant .The patients with rs2239128 genotype who carried the TT allele had higher levels when compared to carriers of no peripheral oedema group（P〈0.05）.Conclusion：The CACNA1C polymorphism rs2239128 TT may be associated with the effectiveness of Peripheral oedema adverse drug reaction of Felodipine.
