摘  要:目的探讨miR-702-3p在纳米二氧化硅(Si O2)致肺纤维化中的生物学作用。方法用Targetscan.org数据库对miR-702-3p调控的靶基因进行预测,通过Gene Ontology和KEGG分析,结合文献检索等方法选择与肺纤维化相关的靶基因为骨形态发生蛋白-7(BMP-7);qRT-PCR与Western blot检测纳米Si O2染尘大鼠60 d组和生理盐水组中大鼠肺组织miR-702-3p、BMP-7 mRNA和BMP-7蛋白的表达水平。结果预测miR-702-3p可能调控的靶基因包括BMP-7共有136个;与生理盐水组比较,染尘组大鼠肺组织中miR-702-3p的表达上调(P<0.05)(与前期高通量测序的结果一致),BMP-7 mRNA的表达水平降低(P<0.05),BMP-7蛋白的表达水平降低(P<0.05)。结论 miR-702-3p与纳米Si O2诱导的肺纤维化相关,其可能通过调控BMP-7等靶基因在肺纤维化中发挥作用。Objective To investigate the biological effects of miR-702-3p on pulmonary fibrosis induced by nanosized SiO2. Methods Potential target genes of miR-702-3p were predicted with database of Targetscan. org. Bone morphogenetic protein 7 ( BMP-7 ) , which associated with pulmonary fibrosis, was filtrated to be the target genes of miR-702-3p for further study with the analysis of Gene Ontology, KEGG and literature retriev- al. The level expression of miR-702-3p, BMP-TmRNA and BMP-7 in pulmonary fibrosis of rat group induced by nano-SiO2 particles and normal saline rat group was detected with quantitative real-time PCR and western blot analysis respectively. Results There were 136 downstream target genes including BMP-7 were regulated by miR-702-3p potentially. The expression of miR-702-3p in rat lung tissue of nano-SiO2 particles group was up- regulated ( P 〈 0.05 ) when compared with the saline group, which was consistent with the result of Illumina Hiseq2000 high throughout sequencing. The expression of BMP-7 mRNA and protein BMP-7 in the lung tissue of nano-SiO2 particles group was both down-regulated (P 〈 0.05 ) when compared with the saline group, the difference was significant statistically. Conclusion MiR-702-3p probably participates in the regulation of pul- monary fibrosis induced by nano-SiO2 particles and BMP-7 may be a target gene of miR-702-3p in regulating pulmonary fibrosis.
