摘　　要：为探索CD44分子在口蹄疫病毒(FMDV)感染中的作用,揭示FMDV的感染机制,为开发阻断FMDV入侵的药物和高效疫苗提供新靶点。采用Western-blot、TCID50检测、激光共聚焦显微镜检测等方法,检测FMDV感染BHK21细胞后CD44的表达水平,以及用抑制剂MβCD抑制CD44表达后对FMDV增殖的影响。Western-blot结果显示,FMDV感染BHK21细胞后,CD44的表达量显著上升(P<0.05);用MβCD抑制内源性CD44表达后,FMDV的增殖水平显著降低(P<0.05)。TCID50检测结果显示,用MβCD抑制CD44表达后,FMDV的表达显著降低(P<0.05)。激光共聚焦结果显示,FMDV感染BHK21细胞后CD44的表达量上升。表明,CD44分子参与FMDV的感染过程,可能作为调控分子直接或间接参与FMDV的入侵过程,仍需要进一步研究。To explore the role of CD44 molecule in foot-and-mouth disease virus（FMDV） infection and to further reveal mechanism of the FMDV infection for the development of inhibitors which block the invasion of FMDV and providing new targeted for effective vaccine.The expression of CD44 in BHK21 cells with FMDV infection was detected in Western-blot,TCID50 and Confocal.In addition,MβCD was introduced as CD44 inhibitor.The result of Western-blot showed that the FMDV infection significantly increased the expression of CD44 in BHK21 cells（P〈0.05）.When repressed the expression of CD44 using the inhibitor MβCD,the expression of FMDV structural proteins was significantly reduced（P 0.05）.In addition,TCID50 assay showed that MβCD treatment decreased the viral titer of FMDV（P〈0.05）.The result of Confocalfurther confirmed that FMDV infection increased the expression of CD44 in BHK21 cells.In conclusion,CD44 possibly functions as a direct molecular or as an indirect molecular to participate in the process of FMDV invasion.Further research is needed to understand its more roles.
