摘 要:目的自主研发制备一种新型的结构合理、使用方便的石蜡组织微阵列手动装置,以解决现有的石蜡组织芯片工具由于结构、组合等原因在实验、研究工作中存在使用不便等问题。方法自主研发制备石蜡组织微阵列手动装置,并利用此装置将62例肝癌组织及匹配的癌旁组织标本制备组织芯片,并对其进行HE染色及AFP、CD34、Ki67和HIF alpha等蛋白质的免疫组织化学实验,分析肝癌组织及癌旁组织的表达差异性。结果对自制组织芯片进行多种蛋白的免疫组织化学检测和分子病理分析结果显示,染色清晰完整;统计分析显示,Ki67和HIF alpha在肝癌组织中的表达量显著高于癌旁组织(P=0.0003,P=0.0144),与文献的报道一致。结论本文自制组织微阵列仪具有较高的实用价值,是生物化学药物分子机制和病理研究应用的有效工具。Objective To make a novel paraffin wax tissue microarray manual device with reasonable structure and handy function,and resolve the inconvenience showed in the usages of existing paraffin tissue microarray tools due to its unreasonable structure,combination and so on in the research work. Methods A novel paraffin wax tissue microarray manual device was made,and 62 cases of hepatocellular carcinoma( HCC) and adjacent tissue array was prepared,then the tissue array was stained by HE,also AFP,CD34,Ki67 and HIF alpha by immunohistochemistry. Results The HCC and adjacent tissue microarray was successfully prepared by self-developed paraffin tissue microarray device. The result of staining is very clear and convenient for molecular pathology analysis. Statistical analysis showed that Ki67 and HIF alpha expression in HCC tissues was significantly higher than that in adjacent tissues( P = 0. 0003,P = 0. 0144),consistent with reports. Conclusion This self-developed tissue microarray manual device with high practical value is an effective tool for the study of the molecular mechanism of biochemical drugs and pathology.
