健康供者骨髓血造血干细胞采集效果分析

摘　　要：目的研究影响骨髓造血干细胞动员及采集的因素。方法方便选取2013年3月—2016年6月期间就诊该院的77名健康供者,经重组人粒细胞集落刺激因子（rh G-CSF）动员后第4天采集骨髓,分析不同健康供者的年龄和性别对骨髓造血干细胞采集的影响以及采集时外周血白细胞数与采集效果的关系。结果 77名供者均成功采集,骨髓采集物单个核细胞数（BM-MNCs）为（4.3±0.43）×10^8/kg,骨髓采集物CD34＋细胞数（BM-CD34^＋）为（1.84±0.30）×10^6/kg,健康供者年龄和性别差异无统计学意义,采集时外周血白细胞数不能预测骨髓采集物中CD34^＋细胞的数量。结论采用5-10μg/kg G-CSF动员,健康供者均可成功采集骨髓干细胞。Objective To study the factors that affect the mobilization and collection of hematopoietic stem cells in bone marrow. Methods Convenient selection ivestigated 77 healthy donors Between March 2013 and June 2016, whose bone marrow were collected at the 4th day after mobilizing by rh G-CSF and analyzed the effects of age and sex on the collection of hematopoietic stem cells in bone marrow of different healthy donors, and the relationship between peripheral blood white blood cell count and collection efficiency. Results Bone marrow hematopoietic stem cells from 77 donors were successfully collected. The amount of total mononuclear cells in the BM mixture grafts were（4.37±0.43）×10^8/kg. The amount of total CD34 ＋ cells in the BM mixture grafts were（1.84 ±0.30） ×10^6/kg. There were no statistical differences in age and sex of healthy donors. The number of white blood cells in peripheral blood can not predict the number of CD34^＋ cells in the bone marrow collection material. Conclusion By mobilization with G-CSF at the dose of 5-10 μg/kg, bone marrow stem cells can be successfully collected from healthy donors.
