摘 要:为探究不定根再生过程中多胺对多胺氧化酶的调控机制,用外源多胺及多胺氧化酶抑制剂处理大豆幼苗下胚轴,检测大豆下胚轴不定根再生情况以及二胺氧化酶基因(Cu AO)表达和酶活性的变化。结果显示,低浓度的腐胺、亚精胺和精胺都显著促进大豆下胚轴不定根的生长,最适浓度分别为1.00 mmol/L、0.25 mmol/L及0.05mmol/L,高浓度多胺则对不定根的生长表现出一定的抑制作用。3种外源多胺最适浓度处理后Cu AO基因表达明显上调,显著高于对照组,且一直维持较高的水平,Cu AO酶活性显著高于对照组。在3种多胺处理中,腐胺处理的效果最好。Cu AO专性抑制剂氨基胍可显著抑制大豆下胚轴不定根的生长,这可能与Cu AO活性受到氨基胍抑制有关。表明,Cu AO基因表达受外源多胺的诱导,多胺促进大豆下胚轴不定根的生长可能与其诱导Cu AO基因表达以及促进酶的活性有关。Soybean [ Glycine max(L. ) Merr. ] seedling hypocotyls were treated with exogenous polyamine and ami- noguanidine to explore the regulating mechanism of polyamine on polyamine oxidase in adventitious root regeneration. The number of adventitious roots,the changes of diamine oxidase gene expression and the enzyme activity were measured. Low concentrations of putrescine ( Put),spermidine (Spd) and spermine (Spm) significantly promoted the growth of soybean hypocotyl adventitious roots,and the optimum concentrations were 1. 00 mmol/L、 0. 25 mmol/L and 0. 05 mmol/L, respec-tively. On the contrary,polyamines at high concentrations showed inhibitory effects. The gene expression level of CuAO wasup-regulated by polyamines and maintained a high level afterwards. Correspondingly,the CuAO activity was gradually in-creased by exogenous polyamines during the regeneration of adventitious roots. Put showed the best effect. In addition,the role of aminoguanidine (A G ,a specific inhibitor of CuAO) was detected. It could significantly inhibit the growth of soybean ad-ventitious root,which might be related to the inhibition of CuAO activity by AG. These results imply that the expression of CuAO gene can be induced by exogenous polyamines, the induction of CuAO gene expression and the improvement of the enzyme activi-ty might have contributed to the promotion of soybean hypocotyl adventitious root growth.
