摘 要:以新农1号狗牙根〔Cynodon dactylon(L.)Per cv.Xinnong No.1〕胚性愈伤为材料,探讨狗牙根离体培养结合甲基磺酸乙酯(EMS)诱变技术体系,利用不同EMS浓度和不同处理时间两个因素对新农1号狗牙根胚性愈伤组织诱变的研究发现:同一处理时间褐化死亡率随EMS浓度的增加呈上升趋势;浓度低于0.4%EMS处理对愈伤致死率无显著影响,而高于0.6%浓度的EMS处理对愈伤致死率有极显著影响。0.8%、1.0%EMS处理2h,0.8%EMS处理4h,0.6%EMS处理6h,愈伤组织致死率都接近半致死剂量。同一处理时间下,愈伤分化率随EMS浓度的升高而呈下降趋势;0.6%EMS处理2h,0.8%EMS处理2h,0.2%EMS处理4h,0.4%EMS处理4h的相对分化率均接近50%。综合愈伤致死率和分化率,新农1号狗牙根愈伤组织诱变最适剂量为0.8%EMS处理2h,此时的致死率为40.56%、相对分化率为54.86%。The embryogenic callus of Cynodon dactylon(L.)Per cv.Xinnong No.1were used as materials,the vitro technology combining with EMS mutageness were explored.The different EMS concentration and different treatment time had mutagenic effects on embryogenic callus mutation of Cynodon dactylon(L.)Per cv.Xinnong No.1.The different concentration and different treatment time were used to research and found:the browning lethal rate increased with the increasing of EMS concentration at the same treatment time;When EMS concentration was lower than 0.4% EMS treatment,there were no significant influence on browning death rate,while the treatment EMS had significant effect on the callus lethal rate,when the treatment with 0.8%,1.0% EMS for 2h,the treatment with 0.8% EMS for 4h,the treatment with0.6% EMS for 6h,the callus lethal rate were close to semi lethal dose.While the same treatment was conducted,the callus differentiation rate decreased with increasing of EMS concencration.The relative differentiation rate of the treatment with 0.6% EMS for 2h,the treatment with 0.8% EMS for 2h,the treatment with 0.2% EMS for 4h,the treatment with 0.4% EMS for 4hwere close to 50%.As far as the comprehensive evaluation with callus lethal rate and differentiation rate,the of suitable dose callus mutation was 0.8%EMS treated for 2h.
