摘  要:为利用变性梯度凝胶电泳(DGGE)技术检测微小牛蜱体内携带立克次氏体的种类,无菌条件下提取微小牛蜱总DNA,以设计的引物RPCS.569P/RPCS.1262n和通用引物RPCS.877P/RPCS.1258GCn扩增立克次氏体gltA基因,DGGE电泳分离,选取条带进行回收克隆测序。结果表明:微小牛蜱之卵、饱血雌成蜱、半饱血雌成蜱、雄成蜱内均只含2种立克次氏体;测序、分析显示分离菌为立克次氏体属斑点热群。说明微小牛蜱具有潜在斑点热病源传播风险。To use the denaturing gradient gel electrophoresis( DGGE) technique to detect the species of Rickettsia carried by Boophilus microplus in vivo,the total DNA of Boophilus microplus was extracted under sterile conditions. The gltA gene of Rickettsia was amplified using designed primers RPCS. 569 P and RPCS. 1262 n,universal primers RPCS. 877 P and RPCS. 1258 GCn. The amplified products were separated with DGGE,and then the selected DGGE bands were recovered,cloned and sequenced. The results showed that only two species of rickettsiae were detected in the four samples from the eggs of Boophilus microplus,full- blood female adult ticks,half- full blood female adult ticks and male adult ticks. The sequencing and analysis showed that the bacteria were spotted fever group rickettsiae. The result indicates that Boophilus microplus has a potential risk of spotted fever transmission.
