摘 要:目的:探讨Roux-en-Y胃转流术对GK大鼠胰腺磷酯酰肌醇3激酶表达的影响。方法:取24只GK大鼠,分手术组(A组,n=8)、假手术组(B组,n=8)、对照组(C组,n=8)。检查各组大鼠术前及术后1、2、4、8周空腹血糖和糖负荷后2h血糖,术后8周处死大鼠取胰腺组织,Western blot测定磷酯酰肌醇3激酶表达量。结果:A组术后1周空腹血糖及糖负荷后2h血糖明显下降,术后8周空腹血糖及糖负荷后2h血糖分别为(6.0±0.3)mmol/L、(7.2±0.5)mmol/L,术前术后比,差异有统计学意义(P<0.05);术后8周,A组大鼠胰腺PI3K表达量显著高于B、C组,差异有统计学意义(P<0.05)。结论:胃转流术可降低GK大鼠血糖,显著增加大鼠胰腺磷酯酰肌醇3激酶的表达量,这可能是该手术治疗2型糖尿病的机制之一。Objective: To investigate the effect of Roux - en - Y gastric bypass on the expression of phosphatidylinositol 3 - kinase in the pancreas of GK rats. Methods: A total of 24 GK rats of 8 weeks old were randomly divided into 3 groups, operation group (A group, n = 8) , sham operation group (B group, n = 8) and control group ( C group, n = 8 ). The blood glucose level of the rats from each groups were measured for the fasting blood glucose level and 2 hours after glucose load, before the operation and at an interval of 1,2, 4, 8 weeks post - operation respectively. Post- operation 8 rats were sacrificed to extract the pancreatic tissues and the Western Blot assay was done to measure the phosphatidylinositol 3 - kinase expression. Results : The fasting blood glucose level of the rats in group A decreased to (7.2 ±0.8) mmol/L from ( 12.5± 1.9) mmol/L after 1 week of operation and the fasting blood glucose level was ( 6.0 ±0.3 ) mmol/L after 8 weeks of operation. The difference between the pre - operative and post - operative blood glucose levels was statistically significant (P 〈 0. 05 ). After 1 weeks of operation, the blood glucose level of 2 hours after glucose load was reduced to (6.9± 0.4 ) mmol/L from ( 14.4 ± 1.7 ) mmol/L. After 8 weeks, the blood glucose level of 2 hours after glueose load was (7.2 ±0.5) mmol/L; there was statistically significant difference between the pre - operative and post - operative blood glucose levels (P 〈 0. 05). At 8 weeks post - operation, the expression of PI3K protein in A group rats were significantly higher than those in group B and group C rats, and the difference was statistically significant (P 〈 0. 05). Conclusions:The gastric bypass surgery ean reduce the blood glucose level of GK rats and signifieantly increase the 3 kinase expression. This may be one of the mechanisms for the treatment of type 2 diabetes mellitus.
