摘  要:目的:研究血清淀粉样蛋白(SA)A1基因多态性与重度肥胖儿童血脂的关系。方法:选择65名重度肥胖儿童作为研究对象,同时选取65名同性别、同年龄的正常体重儿童作为对照组,分别测量他们的身高、体重,计算体重指数,检测血脂(TC、LDL-C、TG)和SAA浓度并进行对比,同时检测四种单核苷酸多态性(SNPs)的基因型,比较两组儿童四种SNPs的分布频率及各基因型间SAA的浓度。结果:重度肥胖儿童SAA血浆浓度和TC、LDL-C、TG一样明显高于正常对照组,rs12218和rs4638289两种SNPs分别频率在重度肥胖儿童中较正常组高,而其它两组SNPs则在两组儿童中无明显差异,但只有rs4638289基因多态性显示与SAA浓度有关。结论:SAA1基因多态性与重度肥胖儿童血脂密切相关。Objective:To study the association of serum amyloid(SA)A1genetic polymorphism with serum lipids in heavy obese children.Methods:A total of 65 heavy obese children and 65 normal children were recruited.Their age and gender ware matched.Height and weight were measured respectively,and BMI was calculated,serum lipids and SAA were tested and compared two groups.Four SNPs of the human SAA1gene(rs12218,rs4638289,rs7131332 and rs11603089)were genotyped by use of polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method.The frequencies of gene and serum SAA level of gene-types were compared between the two groups.Results:Compared with that in controls,circulating SAA levels increased in obese children,so did LDL-C,TG and TC concentration.And obese children showed higher frequency of rs12218 and rs4638289polymorphism.There were no differences between obese and non-obese children for the other 2polymorphisms.Only the rs4638289 polymorphism was related to higher SAA plasma levels.Conclusion:SAA1genetic polymorphism was associated with heavy obesity in Chinese children.
