改良中药复方Formula-3治疗BN大鼠食物过敏模型的疗效及肠道的病理变化

摘  要:目的探讨改良中药复方Formula-3治疗卵清蛋白(ovalbumin,OVA)致敏BN大鼠食物过敏模型的疗效及肠道的病理变化。方法将24只BN大鼠按随机数字表法分为生理盐水对照组、OVA模型组、中药复方Formula-3治疗组和中药复方Formula-3对照组,每组6只。将OVA模型组6只BN大鼠建立OVA模型,均成功建模。前6周,OVA模型组、中药复方Formula-3治疗组BN大鼠均给予OVA混合液灌胃,生理盐水对照组、中药复方Formula-3对照组BN大鼠均给予生理盐水灌胃。第7周开始,中药复方Formula-3治疗组、中药复方Formula-3对照组BN大鼠均给予中药复方灌胃,生理盐水对照组、OVA模型组BN大鼠均给予生理盐水灌胃,同时第7、9、11、13周,OVA模型组、中药复方Formula-3治疗组BN大鼠均给予OVA混合液灌胃。第13周后用100g·L-1 OVA灌胃激发,24h后给予乙醚注射液麻醉,处死,摘除眼球取血。检测各组BN大鼠血清OVA-免疫球蛋白E(IgE)、IL-4的水平,光学显微镜和透射电镜下观察各组BN大鼠肠道组织的病理变化。结果 OVA模型组、中药复方Formula-3治疗组BN大鼠血清OVA-IgE、IL-4水平均明显高于生理盐水对照组、中药复方Formula-3对照组(均P<0.05)。中药复方Formula-3治疗组BN大鼠血清OVA-IgE、IL-4水平均明显低于OVA模型组(均P<0.05)。光学显微镜下观察显示中药复方Formula-3治疗组的肠道炎症程度、肠绒毛破坏明显减轻。透射电镜下观察显示OVA模型组可见肠道上皮细胞消失,微绒毛排列紊乱,细胞器破坏,肥大细胞脱颗粒严重,细胞核固缩、消失。中药复方Formula-3治疗组的肠道上皮细胞排列整齐,微绒毛轻度肿胀,肥大细胞脱颗粒减少,细胞核结构正常。结论改良中药复方Formula-3能够通过保护肠道黏膜屏障和抑制炎症来治疗食物过敏性疾病。Objective To explore the therapeutic efficacy of modified Chinese herbal compound Formula-3 and pathological changes in intestine in ovalbumin(OVA)-induced food allergy Brown Norway(BN) rat model. Methods Twenty-four BN rats were randomly divided into four groups, with 6 rats in each group. Rats in OVA model group and Formula-3 treatment group were given OVA by gavage for 6 weeks. From the seventh week, rats in Formula-3 treatment group and For-mula-3 control group were gavaged with Chinese herbal compound and rats in normal saline (NS) control group and OVA model group were gavaged with NS. Furthermore, rats in OVA model group and Formula-3 treatment group were given OVA at weeks 7,9,11 and 13. After treatment for 13 weeks,rats were anesthetized with ether and sacrificed 24 after excitation with 100 g·L^-1 OVA. Blood samples were collected from the ocular artery after eyeball removal, and serum levels of OVA-IgE and IL-4 were measured. Pathological changes in intestine were observed by optical microscopy and transmission electron microscopy. Results Compared with NS control group and Formula-3 control group, serum levels of OVA-IgE and IL-4 significantly increased in OVA model group and Formula-3 treatment group(P〈0.05). Compared with OVA model group, serum levels of OVA-IgE and IL-4 significantly decreased in Formula-3 treatment group (P〈0.05). Optical microscopy showed that intestinal inflammation and villous damage reduced in Formula-3 treatment group. Transmission electron microscopy showed disappearance of intestinal epithelial cells, disordered arrangement of microvilli, organelle damage, severe degranulation of mast cells, and py- knosis and disappearance of nucleus in OVA model group. However,regular arrangement of epithelial cells,mild swelling of microvilli, decreased degranulation of mast cells and normal structure of nucleus were found in Formula-3 treatment group.
