益气活血通络方含药血清对高糖诱导的大鼠背根神经节细胞NGF、IL-1及TNF-α表达的影响

摘　　要：目的探讨益气活血通络方含药血清对高糖诱导的大鼠背根神经节细胞,神经生长因子、白细胞介素1及肿瘤坏死因子α表达的影响。方法雄性SD大鼠制备含药血清和正常血清,含药血清干预原代培养DRGn,ELISA法检测细胞上清中NGF、IL-1及TNF-α的水平。结果与正常组相比,模型组NGF水平显著下降(P<0.01),IL-1、TNF-α水平明显升高(P<0.05);与模型组相比,含药血清高、中、低剂量组NGF水平均显著升高(P<0.01),TNF-α水平明显降低(P<0.05),高剂量与中剂量组IL-1水平明显降低(P<0.05)。结论益气活血通络方对糖尿病周围神经病变的防治作用的机制可能与其抑制炎症细胞因子的释放,升高NGF含量有关。Objective To explore effects of the expression of NGF,IL-1 and TNF-α of YQHXTL Recipe medicated serum on high glucose induced DRGn. Methods Male SD rats to preparemedicated serum and normal serum.Detect the level of NGF,IL-1 and TNF-α by ELISA.Results Compared with the normal group,the level of NGF of the model group decreased significantly（ P〈0. 01）,IL-1,TNF-α levels increased（ P〈0. 05）;compared with the medicated serum group,NGF levels were significantly increased（ P〈0.01）,IL-1 significantly reduced（ P〈0.05） in high and middle group. TNF-α levels were significantly lower（ P〈0.05）. Conclusions The mechanism of treatment on DPN by YQHXTL Recipe may be related to the inhibition of inflammatory cytokines and increase NGF.
