摘　　要：目的建立红外辅助萃取-高效液相色谱(IRAE-HPLC)法测定丹参药材中丹酚酸B含量的方法。方法 Diamonsil C18柱(150 mm×4.6 mm,5μm),柱温为25℃,以乙腈-0.1%磷酸水溶液(22:78)为流动相;检测波长286 nm,流速1.2 mL·min^(-1)。红外辅助萃取条件的溶剂是20 mL蒸馏水,提取温度90℃,提取时间8 min。结果丹酚酸B在5~100 mg·L^(-1)内线性关系关系良好(r=0.999 1),高、中、低浓度下方法的平均加样回收率为95.26%~108.47%,RSD为1.13%~2.02%(n=3),定量限(RsN=10)为0.5 mg·L^(-1),检测限(RsN=3)为0.15 mg·L^(-1)。用该方法实际测得3批丹参药材中丹酚酸B的含量可达3.9%~4.0%。结论该方法简便、快速、准确、可靠、安全、环保,适用于测定丹参药材中丹酚酸B的含量。AIM To develop an infrared-assisted extraction method coupled with HPLC （IRAE-HPLC） method for the determination of salvia acid B in Salvia miltiorrhizae. METHODS Salvia acid B was determined by HPLC on a Diamonsil Cls column （150 mm × 4.6 mm, 5μm） at 25℃ with acetonitrile- 0.1% phosphoric acid aqueous （22：78） as the mobile phase. Detection wavelength was 286 nm. The flow rate was 1.2 mL· min^- 1. Salvia acid B was extracted by IRAE with 20 mL deionized water using extraction condition of 90℃ and 8 rain. RESULTS The standard curves for salvia acid B were linear over the range of 5 - 100 mg·L^- 1 （ r = 0. 999 1 ）. The average recoveries of salvia acid B were between 95.26% and 108.47% with RSD from 1.13% to 2.02% （ n = 3）, the limit of quantity was 0.5 mg·L^- 1and the limit of detection was 0.15mg·L^- 1. The method was applied for analyzing salvia acid B in Salvia miltiorrhizae successfully, the average content of salvia acid B in three practical batch samples were 3.9%. CONCLUSION The method is simple, accurate and with high efficiency. It is adapted to determine and analyze salvia acid B in Salvia miltiorrhizae.
