Effects of Ad-HIF-1α-564/402 gene transfer on the myocardial ischemi injury and Na^+-K^+ ATP enzyme in rats

摘  要:目的:研究重组腺病毒低氧诱导因子-1α双突变体基因对心肌梗死大鼠缺血心肌损伤程度及Na^+-K^+ATP酶的影响.方法:36只雄性SD大鼠建立急性心肌梗死模型,随机分成3组,分别为生理盐水组(Saline组,n=12)、腺病毒空载体组(Ad-Null组,n=12)、Ad-HIF-1α-564/402组(n=12).在术后即刻分别肌注生理盐水、AdNull、Ad-HIF-1α-564/402 0.1 m L,病毒滴度1.0×108PFU.术后7 d,记录大鼠24 h饮食量、饮水量及体质量,观察大鼠的精神及运动状况.检测血清中肌酸激酶同工酶、肌钙蛋白I、B型利钠肽浓度.进行血流动力学检测,测量左室收缩压、左室舒张末压、左室最大上升/下降速率.采用分光光度计法检测心肌梗死大鼠心肌Na^+-K^+ATP酶水平.结果:基因转染后7 d,3组大鼠饮食量、饮水量、体质量无统计学差异.Ad-HIF-1α-564/402组大鼠较Saline组、AdNull组反应更为灵敏,活动耐量增加.CK-Mb、c Tn I、BNP血清浓度在Ad-HIF-1α564/402组均较其他两组为低,3组间有统计学差异(P<0.05).Ad-HIF-1α-564/402组大鼠LVSP、±LVdp/dtmax明显高于Saline组、Ad-Null组(P<0.05).Ad-HIF-1α-564/402组大鼠LVEDP明显高于Saline组、Ad-Null组(P<0.05),上述指标Saline组、Ad-Null组均无统计学差异(P>0.05).分光光度计结果显示:Na^+-K^+ATP酶水平3组间无统计学差异(P>0.05).结论:AdHIF-1α-564/402能减轻心肌细胞梗死程度,保护心脏功能,对Na^+-K^+ATP酶水平无明显影响.Aim: To investigate the effect of the recombinant adenovirus containing the 564/402-point- mutant HIF-la gene (Ad-HIF-1α-564/402) on the myocardial ischemia injury and Na^+-K^+ATP en-zyme in rats. Methods 36 male SD rats of acute myocardial infarction model were randomly divided into 3 groups: Saline group (n = 12) , Ad-Null group (n = 12) , and the Ad-HIF-1α-564/402 group (n= 12) . The rats were given saline, Ad-Null, or Ad-HIF-1α-Trip 0. 5 mL (4 × 10^8PFU) respectively by in-tramuscular injection. 7 days after gene-transfection, the mass of food and the volume of water per 24 hours were calculated. The weight and exercise of the rats were recorded. The level of CK-Mb, cTnI and BNP in serum were also detected. Hemodynamic changes including left ventricular systolic pressure (LVSP) , left ventricular diastolic pressure (LVEDP) , and the maximum rate left ventricular pressure rised or decreased (± LVdp/dtmax) were recorded. Spectrophotometry was chosen to detect the level of Na^+-K^+ATP enzyme in the ischemic myocardial tissue. Results: On the 7th day after gene-transfection, there were no significant differences of the food and water-intaken, as well as the weight of the SD rats a-mong the three groups. The rats of Ad-HIF-1α-564/402 group were more sensitive and activity tolerance than the other two groups. Compared with Saline and Ad-null groups, the LVSP and ± LVdp/dtmax of rats in Ad-HIF-±-564/402 group increased (P 〈0. 05) , whereas LVEDP decreased (P 〈0. 05) . The lev-els of CK-Mb,cTnI and BNP in the Ad-HIF-±-564/402 group were the lowest among the three groups (P 〈 0. 05 ) . The levels of Na^+-K^+ATP enzyme detected by spectrophotometry had no significant differ-ences among the three groups. Conclusion: Ad-HIF-1α-564/402 can cut down the degree of myocardial infarction, protect the heart function without having influence on Na^+-K^+ATP enzyme.
