自拟消癖汤治疗雄性大鼠乳腺增生实验研究

摘　　要：目的观察自拟消癖汤对雄性大鼠乳腺增生的治疗效果,以指导男性乳房肥大症的治疗作用研究。方法腹腔注射给予苯甲酸雌二醇(1.0 mg/kg),1次/d,连续20 d,以复制雄性大鼠乳腺增生模型。60只SPF级Wistar雄性大鼠随机均分为正常对照(等溶生理盐水)组、模型(等溶生理盐水)组、三苯氧胺(1.8 mg/kg)组、自拟消癖汤低、中、高(2.5、5、7.5 m L/kg)剂量组,各组大鼠均于复制模型结束后腹腔注射给药,2次/d,连续30 d。采用酶联免疫吸附(ELISA)法测定大鼠血清雌二醇(E_2)、睾酮(T)的水平,观察大鼠乳腺组织结构变化,对比评价疗效。结果与正常对照组比较,模型组乳腺增生明显,血清E_2增加,血清T含量减少;差异均有统计学意义(P<0.05)。与模型组比较,三苯氧胺组与自拟消癖汤低、中、高剂量组大鼠乳腺增生均有不同程度减轻,血清E_2水平降低,血清T水平升高,差异均有统计学意义(P<0.05);与三苯氧胺组比较,自拟消癖汤高剂量组大鼠血清E_2水平降低,血清T水平升高,乳腺组织增生程度明显减轻,差异均有统计学意义(P<0.05)。结论自拟消癖汤对雄性大鼠乳腺增生有明显的抑制作用,疗效显著。Objective To observe the therapeutic efficacy of self designed Xiaopi decoction on hyperplasia of mammary gland in the male rats,to guide the therapeutic study on treatment of male breast hypertrophy. Methods The formation of mammary gland hyperplasia by ip estradiol benzoate（1. 0mg / kg）,twice a day for 20 d. Sixty SPF Wister male rats were divided into normal control group（ isovolumic normal saline）,model group（ isovolumic normal saline）,tamoxifen group（1. 8 mg / kg）,and self designed xiaopi decoction low,medium and high dose groups（2. 5,5,7. 5 m L / kg）. The groups were given ig administration,once a day for 30 d. The concentrations of rats serum estradiol（E_2）and testosterone（T）were detected by ELISA. Observe the changes of mammary gland organization structure and compare the curative effects. Results Compared with the normal control group,model group,the concentrations of serum E_2 was increased and T was reduced. The differences were statistically significant（ P 〈0. 05）. Compared with the model group,the degree of mammary gland hyperplasia in rats have reduced which in tamoxifen group and self designed Xiaopi decoction low,medium and high dose groups to some extent,the concentrations of serum E_2 was de-creased and Serum T was increased,there was statistical significant difference（ P 〈0. 05）. Compared with the tamoxifen group,the concentrations of serum E_2 was decreased and Serum T was increased in self designed Xiaopi decoction high dose groups,the degree of mammary gland hyperplasia in rats have reduced obviously,there was statistical significant difference（ P 〈0. 05）. Conclusion The self designed Xiaopi decoction has significant curative effect on hyperplasia of mammary gland in the male rats.
