摘 要:目的探讨红色毛癣菌毒力因子的相关基因金属蛋白酶(metalloprotease,MEP)和几丁质合酶(chitin synthase,CHS)mRNA在甲真菌病体外模型中的变化及其致病作用。方法构建甲真菌病体外模型,随机分为实验组和对照组(未加甲板),分别于培养的第1,2,3,4和5周时提取红色毛癣菌总RNA,采用实时荧光定量PCR检测不同时间点MEP mRNA和CHS mRNA的表达情况,并进行统计学分析。结果对照组MEP mRNA和CHS mRNA的表达极低,实验组在培养时间内MEP mRNA和CHS mRNA的表达呈增加趋势。而CHS mRNA在第4周时表达已达高峰,此后逐渐下降,但仍高于对照组。在各时间点与对照组相比,差异均有统计学意义(P均<0.05)。结论金属蛋白酶和几丁质合酶在甲真菌病体外模型中表达较高,其在甲真菌病的致病中可能起重要作用。Objective To investigate the genes expression of virulenee from Triehophyton rubrum and pathological role of metalloprotease (MEP) and chitin synthase (CHS) mRNA in vitro onyehomycosis model. Methods To- tal RNA of Triehophyton rubrum in the treated group and control group ( non-infected nails) was extracted from cultured onychomyeosis model at different time points ( 1 w, 2w, 3w,4w and 5w). The expression of MEP mRNA and CHS mRNA at different time points were detected by real-time fluorescence quantitative PCR method, respectively, and performed statistical analysis. Results The expression of MEP mRNA and CHS mRNA was very low in eontrol group, but they showed an increasing trend in treated group. The expression of CHS mRNA began to increase at 1 w,2w and 3w, and reached a peak at 4w, then gradually decreased, but still higher than control group. Compared to the control group, the expression leves of MEP mRNA and CHS mRNA at each time points in the treated group were statistically signifieantly ( P 〈 0. 05 ). Conclusion In vitro onyehomyeosis model, the expression levels of MEP mRNA and CHS mRNA are higher in euhured keratin from nail, it suggests that MEP and CHS may play an important role in the pathogenesis of onyehomyeosis.
